Metrorrhagia refers to the abnormal uterine bleeding in the non menstrual period.It mainly occurs among teenagers and the menopausal transition period female.According to the physiological and pathological characteristics,Liver and Kidney Yin deficiency metrorrhagia is a syndrome containing abnormal uterine bleeding along with soreness and weakness of waist and knees,dizziness and tinnitus;anxiety and irritability,palms and soles,tidal fever and night sweating;red tongue,the tongue coating less,string thin and rapid pulse.At present,the common treatment of western medicine is sex hormones,but the long-term application of estrogen and progesterone has severe side effects.While traditional Chinese medicine has significant advantages in the treatment of such diseases.Erzhiwan is composed of fructus ligustri lucidi and ecliptae herba,which has the effect on tonifying liver and kidney,nourishing Yin and hemostasis,and has definite curative effect in the treatment of Liver-Kidney Yin deficiency metrorrhagia.So far.there is still no research on the material basis and molecular mechanism of Erzhiwan in the treatment of this disease.Recently,network pharmacology has been widely used in the research of traditional Chinese medicine.Its systematic and comprehensive observation of the intervention and influence of drugs on the disease network is just suitable for the overall study of the 'multi-component-multi-target-multi-pathway,mechanism of traditional Chinese medieine.In this study,the material basis and molecular mechanism of Erzhiwan for the treatment of Liver-Kidney Yin deficiency metrorrhagia were discussed by combining literature review and network pharmacology.Objective:To investigate the molecular mechanism of Erzhiwan in treating Liver-Kidney Yin deficiency metrorrhagia,network pharmacological method was used to predict the key targets and signal pathways.Based on literature review of traditional Chinese and western medicine and the experimental research progress of Erzhiwan in the treatment of this disease,this study screened the major active components of Erzhiwan and constructed the active compounds-targets network by using the network pharmacology method.Methods:TCMSP was used to screen the active compounds of Erzhiwan,the key targets of active compounds were collected from multiple databases and confirmed by literature mining.Moreover,protein-protein interaction network(PPI)was built by using the String database.This study used DAVID to analyze Gene Ontology and the pathway of target proteins.Then,the compounds-targets network and the analysis were carried out by Cytoscape software.In addition,the molecular function and biological pathway of the key targets were also performed by DAVID database with P<0.05 and FDR<0.05.Results and conclusion:10 active compounds such as quercetin,luteoline,apigenin and kaempferol were screened out by ADME calculation method.Those compounds acted on the 92 key targets such as STAT3,IL-6,TNF and IL-4.Furthermore,Erzhiwan regulated 29 metrorrhagia-related pathways such as VEGF pathway,progesterone-mediated oocyte maturation.Erzhiwan achieved the result of haemostatic,hormonal regulation by affecting the vascular function,neuroendocrine,glucose-lipid metabolism intervention,inflammation and immune regulation.
