| Objective:To understand the rule of drug use for recurrent spontaneous abortion in the past 10 years and to study the effect of traditional Chinese medicine on the intervention of recurrent abortion in mice with unknown causes and its effect on the balance of Treg/Th17 cells and related factors,To explore the mechanism of the traditional Chinese medicine nourishing kidney and strengthening spleen for the recurrence spontaneous abortion with unknown causes.Methods:1.In terms of traditional Chinese medicine,by reading and collating ancient literature,learning the etiology,pathogenesis,and classic and therapeutic prescriptions of recurrent spontaneous abortion,the research and treatment of the disease in various schools of modern Chinese medicine,and the treatment of the disease by modern Chinese medicine;Western medicine,through reading the Chinese and foreign literature of the disease,understand the pathogenesis of the disease and treatment measures,learning the latest progress.2.In the past 10 years,we have selected the related literature on the treatment of recurrent abortion in Chinese knowledge resources database and Wanfang data knowledge service platform,and the Weipu database has published the relevant literature on the treatment of recurrent abortion.The clinical efficacy observation,and the medication route is oral,and a total of 211 first prescriptions have been obtained after finishing.Based on the analysis and data mining of 211 prescription materials,the paper discussed the rule,core combination and alternative method of treatment for recurrent abortion.3.The experiment was divided into 4 groups:CBA/J female rats x BALB/c male mice were normal pregnancy models(normal pregnancy groups),and CBA/J female mice x DBA/2 male mice were abortion models.The abortion model was randomly divided into 3 groups:Chinese medicine group 10,digestone group 10,URSA group 10.All pregnant rats began to pour their stomachs on the first day of pregnancy,and were executed on the 14th day of pregnancy.The following experiments were conducted to collect peripheral blood and spleen:1 Calculate the embryo loss rate;The concentrations of IL-6,IL-10,IL-17 and TGF-β were detected by ELISA.3 Flow cytometry to detect the expression of mouse spleen Treg/CD4+and Th 17/CD4+cells.Results:1.Compared with URSA group,the rate of embryo loss in traditional Chinese medicine group,diprogestone group,and normal pregnancy group decreased significantly,and the difference was statistically significant,P<0.05;The rate of embryo loss decreased in Chinese medicine group,but the difference was not statistically significant,P>0.05;Compared with the normal pregnancy group,the rate of embryo loss in the traditional Chinese medicine group increased,and the difference was statistically significant,P<0.05.2.Compared with URSA group,the contents of IL-6 and IL-17 cytokines in traditional Chinese medicine group,diprogestone group,and normal pregnancy group were significantly reduced,and the difference was statistically significant,P<0.05;The increase of IL-6 cytokines and the decrease of IL-17 cytokines in Chinese traditional medicine group were not statistically significant compared with those in diprogestone group,P>0.05;Compared with the normal pregnancy group,the contents of IL-6 and IL-17 cytokines in the traditional Chinese medicine group increased,and the differences were statistically significant,P<0.05.3.Compared with URSA group,the content of IL-10 cytokines in Chinese traditional medicine group,diprogestone group,and normal pregnancy group increased significantly,and the difference was statistically significant.P<0.05;The content of TGF-β cytokines in Chinese traditional medicine group and diprogestone group increased,but the difference was not statistically significant,P>0.05;The TGF-β cytokine content in normal pregnancy group was significantly increased,and the difference was statistically significant,P<0.05;The content of IL-10 and TGF-β cytokines in traditional Chinese medicine group increased,but the difference was not statistically significant,P>0.05;Compared with the normal pregnancy group,the content of IL-10 and TGF-β cytokines in the traditional Chinese medicine group decreased,and the difference was statistically significant,P<0.05.4.Compared with URSA group,the ratio of spleen CD4 + CD25 +Treg cells in the traditional Chinese medicine group,diprogestone group,and normal pregnancy group increased significantly,and the difference was statistically significant,P<0.05;The expression level of Th17 cells in Chinese traditional medicine group,diazoprogesterone group,and normal pregnancy group was significantly reduced,and the difference was statistically significant,P<0.05;Compared with the diazoprogesterone group,the ratio of CD4+CD25+Treg cells in the Chinese medicine group increased,and the expression level of Th17 cells decreased,but the difference was not statistically significant,P>0.05;Compared with the normal pregnancy group,the ratio of CD4+CD25+ Treg cells in the Chinese medicine group decreased,and the expression level of Th17 cells increased,and the differences were statistically significant,P<0.05.5.Based on the analysis of TCM inheritance support platform,it is concluded that the deficiency tonic drugs are the most frequently used in TCM treatment of recurrent abortion,followed by heat-clearing drugs and qi regulating drugs.The medicinal properties were mainly warm,smooth,sweet,bitter and bitter,mostly attributed to liver,kidney and spleen meridians.6.Based on the traditional Chinese medicine inheritance and assistance platform,10 core combinations for the treatment of recurrent abortion were discovered,and 5 new prescriptions for the treatment of recurrent abortion were obtained through the core combination analysis.Conclusion:(1)The method of bushing kidney and strengthening spleen was used to intervene the mouse model of recurrent abortion with unknown causes.It was found that it could significantly reduce the embryo loss rate and improve the pregnancy outcome.(2)Chinese traditional medicine nourishing kidney and spleen to reduce IL-6 and IL-17 levels in the serum of pregnant mice and increase IL-10 and TGF-β levels.This may be part of the mechanism for its inhibition of URSA abortion..(3)The traditional Chinese medicine nourishing spleen method increases the proportion of CD4+CD25+Tree in the spleen of the pregnant mouse to CD4+T cells,and reduces the proportion of Th17 cells in the spleen of the pregnant mouse.This may be part of the mechanism by which URSA acts as an inhibitory effect on abortion.(4)the pathogenesis of unexplained recurrent abortion may be related to decreased il-10 and TGF-β,increased il-6 and il-17,decreased proportion of CD4+CD25+Treg in CD4+T cells and increased Th17 cells.(5)Chinese medicine treats recurrent abortion to make up for deficiency medicine,followed by antipyretic medicine and Qi medicine.The medicinal taste is mainly warm,flat,sweet,bitter,and Xin,mostly attributed to the liver,kidney,and spleen.(6)Based on the software of the Chinese medicine inheritance support system(V2.5),the input,management and analysis of prescriptions are completed.It is of great significance for the study of the rules of traditional Chinese medicine groups and the discovery of new prescriptions,and it has a certain guiding role in the clinical treatment of recurrent abortion.(7)The traditional Chinese medicine treatment of
