Objective Through the intervention of five elements music therapy(Gong tune music)in aging model mice,the effects of Gong tune music on learning and memory ability,acetylcholinesterase(Ach E),and nitric oxide(NO)in brain area of aging model mice were observed,and the value of palace mode music in the treatment of Alzheimer's disease was discussed.MethodThirty Kunming male mice aged 3 months were randomly divided into blank control group,music group and model control group with 10 mice in each group.The aging model was established by intraperitoneal injection of D-galactose 120 mg/kg daily for60 days.The music group played music for 12 hours every day from 19:00 to 7:00while establishing the aging model.The repertoire was Gong tune music.When listening to music,the mice were placed in a quiet and fixed room,closed doors and windows,drew curtains,and placed in a fixed position each time.The distance between the sound(SANSUI sound)and the cage was 0.5m,and the decibel was40-50 d B.The mice in the blank control group were intraperitoneally injected with normal saline every day.All mice in the three groups ate and drank water normally,following the guidance of animal ethics.After 60 days of model building and music intervention,the mice were tested by Morris water maze positioning navigation test and space exploration test,and their learning and memory abilities were evaluated by the percentage of the original platform and swimming speed.After Morris water maze test was completed,the brain tissues of three groups of mice were decapitated and ACh E activity and NO content in brain tissues were detected according to the kit instructions.All experimental data were analyzed and processed by SPSS22 statistical software.The results were expressed as mean ±standard deviation and the test level was a=0.05.Result1.The results of positioning navigation test showed that there was no significant difference in the escape latency of each group on the first day(P>0.05);the escape latency of the model control group was longer than that of the music group and the blank control group from the second day to the sixth day.There was statistical difference in the escape latency of the model control group compared with the blank control group on the third,fifth and sixth day.(P<0.05);compared with the music group,the escape latency of the fourth and sixth day in the model control group had significant difference(P<0.05);compared with the blank control group,the escape latency of the fifth day in the music group had significant difference(P<0.05).In the positioning navigation test,the swimming speed of the model control group was lower than that of the music group and the blank control group from the second day to the sixth day;the swimming speed of the model control group was significantly different from that of the blank control group on the fifth day(P<0.05);the swimming speed of the model group was significantly different from that of the music group on the fourth and sixth day(P<0.05).There was no significant difference in swimming speed between the blank control group and the music group(P>0.05).2.The results of space exploration experiment showed that the percentage of platforms passing through the blank control group was the largest,while the percentage of platforms passing through the model control group was the smallest;there was no statistical difference between the music group and the blank control group and the model control group(P>0.05);there was a unified difference between the model control group and the blank control group in the percentage of platforms passing through the model control group(P<0.05).3.The results of biochemical indicators showed that the content of NO and ACHE activity in brain tissue of blank control group were the least,and that of model control group was the most.There was no significant difference in NO content and ACHE activity between the blank control group and the music group(P>0.05).Compared with the blank control group and the music group,there were significant differences in NO content and ACHE activity between the model control group and the blank control group(P<0.05).conclusion1.Gong tune music therapy can improve the learning and memory ability of aging model mice,reduce NO content and ACHE activity in brain tissue.2.Improving the learning and memory ability of aging model mice by Gong tune music therapy may be related to the decrease of NO content and ACHE activity in brain tissue.
