Objective: To investigate the effects of electroacupuncture on limb paralysis in stroke patients by electroacupuncture on the effects of GABAa receptors and synaptic plasticity-related proteins in hippitocampus of rats wh stroke.Methods: Seventy-seven SD male healthy rats(SPF grade)were randomly divided into a blank group,a sham operation group and a model reserve group according to a random number table,of which 9 were blank group and sham operation group.The modified MACO suture method and internal capsule injection NMDA receptor method was used for modeling.The 18 rats that were successfully modeled were randomly divided into two groups: model group and electroacupuncture group,with 9 rats in each group.After the successful modeling,the electroacupuncture group was treated with electroacupuncture at the bilateral Quchi and bilateral Yanglingquan,30 minutes each time,once a day for 5 days;Using the Zea longa nerve damage score for neurological function scores.Assessment of muscle tone using a modified Ashworth muscle tone score;ATPase staining was used to detect the proportion of type I and type II fibers in the biceps and quadriceps of the affected rats.ELISA was used to detect plasma VEGF and PAO levels.The expressions of GABAa receptor,SYN and PSD95 mRNA in rat hippocampus were detected by RT-PCR.The relative expression of GABAa receptor,SYN and PSD95 in rat hippocampus was detected by Western Blot.Results: 1.Zea longa nerve damage score,Ashworth muscle tone score :Compared with the blank group before treatment,there was no statistical significance in the sham operation group(P>0.05),and the model group increased,which was statistically significant(P<0.01),indicating that the stroke limb paralysis model was successfully prepared.Compared with the model group,the electroacupuncture group decreased after treatment,and the difference was statistically significant(P<0.05),indicating that electroacupuncture can improve the neurological function and muscle tone of rats.2.Proportion of type I and type II fibers in the biceps and quadriceps of the affected rats :Under alkali staining(pH 10.7),type I muscle fibers exhibited a light gray color,and type II muscle fibers exhibited a dark brown color.The muscle fibers of the biceps and quadriceps of each group did not show a checkerboard pattern.The type I muscle fibers and type II fibers were scattered,and some muscle fibers were interrupted,and autolysis was observed.3.The content of VEGF and PAO in rat serum: Compared with the blank group,there was no difference in the sham operation group(P>0.05),the VEGF of the model group decreased significantly(P<0.01),and the PAO content decreased(P<0.05).Compared with the model group,there was no difference in the electroacupuncture group(P> 0.05).There was no significant difference in the proportion of type I muscle fibers between groups(P>0.05).4.Expression of GABAa receptor mRNA,SYN mRNA and PSD95 mRNA in hippocampus of rats:Compared with the blank group,there was no difference in the sham operation group(P>0.05).Compared with the blank group,the GABAa receptor,SYN and PSD95 mRNA in the model group decreased significantly(P<0.01),indicating that the stroke limb paralysis model was successfully modeled.Compared with the model group,the GABAa receptor,SYN and PSD95 mRNA were increased in the electroacupuncture group,and the difference was statistically significant(P<0.05).5.Expression of GABAa receptor,SYN and PSD95 protein in hippocampus of rats:Compared with the blank group,there was no difference in the sham operation group(P>0.05).Compared with the blank group,the GABAa receptor,SYN and PSD95 protein in the model group decreased significantly,the differences statistically significant(P<0.01),indicating that the stroke limb paralysis model was successfully modeled.Compared with the model group,the GABAa receptor,SYN and PSD95 protein were increased in the electroacupuncture group,and the difference was statistically significant(P<0.05).Conclusion: 1.Electroacupuncture can improve the neurological function and muscle tone of stroke rats with limb paralysis;2.Electroacupuncture can regulate the expression of GABAa receptor,SYN,PSD95 mRNA and protein in hippocampus of rats with stroke and limb paralysis.It is suggested that electroacupuncture can alleviate the mechanism of limb paralysis in stroke by regulating the GABAa receptor and synaptic plasticity related proteins SYN and PSD95 in the hippocampus.
