The Effect Of Achyranthes Bidentata Polysaccharide On The Expression Levels Of Fadd, Beclin-1 And LC3Ⅱ/I In Human KOA Chondrocytes | | Posted on:2020-01-12 | Degree:Master | Type:Thesis | | Country:China | Candidate:C C Li | Full Text:PDF | | GTID:2434330575968162 | Subject:Fractures of TCM science | | Abstract/Summary: | | | Background: The pathological changes of KOA are mainly degenerative change s of chondrocyte,which are manifested by increased apoptosis of chondrocyte.Autophagy is an intrinsic protective mechanism for KOA chondrocytes to repair themselves.With the development of KOA,the autophagy level of chondrocyte s will be inhibited.Relevant studies have shown that improving the autophagy l evel of chondrocytes can decrease the incidence of apoptosis,and then play a p rotective role on chondrocytes,which provides a direction for the prevention an d treatment of KOA.AIM: To investigate the effects of Achyranthes bidentata polysaccharides on the expression of Fadd,Beclin-1 and LC3Ⅱ/Ⅰ in human knee osteoarthritic chondro cytes by using different concentrations of Achyranthes bidentata polysaccharides.Methods: Chondrocytes from KOA patients were cultured and passaged.The thi rd generation chondrocytes were divided into six groups.The blank group was20%FBS+DMEM / F12,and the control group was IL-1beta+20%FBS+ DMEM/F12.The experimental group was divided into 25,50,100 and 200 mg/L Achyr anthes bidentata polysaccharide+IL-1beta+20%FBS+ DMEM/F12.The concentrati on gradient of the experimental group was 25,50,100 and 200 mg/L Achyrant hes+IL-1beta+20%FBS+ DMEM/F12;24 hours after intervention,the expression levels of Beclin-1,LC3Ⅱ/Ⅰ and FADD of chondrocyte were detected by western-b lot.Results: Achyranthes bidentata polysaccharide inhibited the expression of Fadd i n chondrocytes to a certain extent,and promoted the expression of Beclin-1 and LC3Ⅱ/Ⅰ.Compared with the blank group,the expression of Fadd was increased,and the expression of Beclin-1 and LC3Ⅱ was decreased.The expression level of LC3Ⅰ was increased,that is,the ratio of LC3Ⅱ/Ⅰ was decreased,P<0.05,the difference was statistically significant.Compared with the control group,the ex pression of Fadd was generally decreased.The trend of Beclin-1 and LC3Ⅱ was increased,the expression of LC3Ⅰ was decreased,and the ratio of LC3Ⅱ/Ⅰ was increasing(P<0.05).The expression of Fadd was significantly decreased in the concentration of 100mg/L Achyranthes polysaccharides and the concentration of25mg/L Achyranthes polysaccharides.The expression of Beclin-1 and LC3Ⅱ/Ⅰ wa s up,P<0.05 The difference was statistically significant.There was no significan t difference in the expression of each protein compared with the 100 mg/L Ach yranthes polysaccharide concentration group.CONCLUSION: Achyranthes bidentata polysaccharide can improve the autophag y level of KOA chondrocyte and inhibit the apoptosis of KOA chondrocyte,whi ch may be through promoting the occurrence of chondrocyte autophagy to inhib it the apoptosis of chondrocyte. | | Keywords/Search Tags: | KOA, chondrocyte, cell culture, Achyranthes bidentata polysaccharide, Fadd, Beclin-1, LC3Ⅱ/Ⅰ | | Related items |
