| ObjectIn this study,ovariectomized female mice were used as models to explore the preventive and therapeutic effects of water decoction of root bark of Aralia echinocaulis Hand.-Mazz.on osteoporosis in terms of serology,bone morphometry and bone microstructural changes.MethodsEighty 15-week-old SPF female mice were randomly divided into sham-operated group(16 mice)and ovariectomized group(64 mice).Ovariectomized group removed bilateral ovaries.Sham-operated group removed the same size of fat beside ovaries.Continuous injection of 100,000 units of penicillin injection for 7 days after operation.Vaginal pictures of mice were observed to detect estrous cycle,and animals with incomplete estrous cycle were screened for further administration.Forty rats were randomly divided into model group(OVX,n=10),alendronate sodium group(AST,n=10),Xianlinggubao group(XL,n=10),water decoction group(CLB,n = 10)and sham operation group as control group(CG,n=10).The mice in each group were fed with distilled water of equal volume.The weight of mice was measured weekly.After 12 weeks,the mice were sampled.Serum calcium,phosphorus,estradiol,osteocalcin and bone alkaline phosphatase levels were measured.The weight of uterus and femur of mice was measured.Three mice in each group were randomly selected for bone morphometry and microstructure detection.ResultsThe weight of mice in CG group was significantly lower than that in OVX group at 1 and 2 weeks after administration(p<0.05).At 10 weeks after administration,the weight of mice in CG group was significantly lower than that in OVX group(p<0.05).At 12 weeks after administration,the weight of mice in AST group was significantly higher than that in control group(p<0.05).The uterine coefficient of mice in OVX group,AST group,XL group and CLB group was significantly lower than that of mice in OVX group,AST group,XL group and CLB group.CG group(p<0.05);femoral weight coefficient of mice:12 weeks after administration,femoral bone weight coefficient of OVX group,AST group,XL group and CLB group were significantly lower than CG group(p<0.05);serum calcium and phosphorus:12 weeks after administration,there was no significant difference between each group and CG group;serum estradiol:compared with CG group,serum estradiol level of OVX group was significantly lower(p<0.05);Bone morphometry of mice:compared with CG group,TBV,TFS,AFS,MAR,BFR,OSW and mAR in OVX group decreased;compared with OVX group,TFS,AFS,AFS,MAR,BFR,OSW and mAR in AST group,XL group and CLB group decreased;compared with OVX group,TFS,AFS,MAR and BFR in AST group,XL group and CLB group decreased significantly.The total volume,mineralization percentage and average mineralization density of femoral callus in OVX group decreased compared with CG group(p<0.05);compared with OVX group,the total volume,mineralization percentage and average mineralization density of femoral callus in CLB group,XL group and AST group increased significantly(p<0.05).Conclusion1.The water decoction of the root bark of Aralia echinocaulis Hand.-Mazz.could improve the serum estradiol level in ovariectomized mice,and had estrogen-like effect;2.The decoction of the root bark of Aralia echinocaulis Hand.-Mazz.could decrease the level of serum bone alkaline phosphatase and improve bone turnover in ovariectomized mice;3.The decoction of the root bark of Aralia echinocaulis Hand.-Mazz.may regulate bone mass,bone formation and bone resorption by improving the number of osteoblasts and osteoclasts;4.The decoction of root bark of Aralia echinocaulis can promote bone mineralization during bone reconstruction and reduce the possibility of micro-cancellous bone fracture. |
