Objective Via studying on the chemical constituents of non-alkaloids active fractionfrom the root of Aconitum flavum Hand.-Mazz. and screening of monomer with T cellinhabition activity.This work provided the experimental basis for further in-depthstudy on anti rheumatoid arthritis mechanism of Aconitum flavum Hand.-Mazz..Methods1. The root of Aconitum flavum Hand.-Mazz. was extracted by Solventmethod,and enriched by Macroporous Adsorption Resin and Cation Exchange Resin.2.With multiple methods,chemical compounds of Aconitum flavum Hand.-Mazz.active fraction were isolate by silicagel column,ODS reversed phase column,MCIphase column,gel column and TLC.Structure of compounds were identificated bymodern spectroscopic techniques.3.MTT assay were used to observe the effect of non-alkaloids active fraction ofAconitum flavum Hand.-Mazz.,and mono-compound onJurkatcellproliferationResults1.Crude extract0.8kg was extracted from the Herbs10kg.We obtained93.3gfrom30%alcohol by Macroporous Adsorption Resin,yield of11.6%;46.7gnon-alkaloid fraction by Cation Exchange Resin,yield of50.1%.2. There are12monomeric compounds,and identified6monomericcompounds.CompoundⅠ:Di-(2-ethylhexyl)phthalate,compoundⅡ:(2E)-3-[4-(β-D-glucopyranoside)-phenylacrylic]-acid,compoundⅢ:pentadecanoic acid,compoundⅣ:linarin,compoundⅤ:β-sitosterol,compoundⅥ:5-hydroxymethyl furfural. 3.Non-alkaloids active fraction of Aconitum flavum Hand.-Mazz. had significanteffect on inhibiting the proliferation of Jurkat cells at4mg/ml,2mg/ml and1mg/mlconcentration(P<0.05),respectively;five concentration of compound Ⅰhad significanteffect on inhibiting the proliferation of Jurkat cells(P<0.05);compoundⅡ hadsignificant effect on inhibiting the proliferation of Jurkat cells in a concentration of2.6mg/ml,1.3mg/ml,0.75mg/ml,0.375mg/ml (P<0.05); compound Ⅲ hadsignificant effect on inhibiting the proliferation of Jurkat cells at1.98mg/ml (P<0.05); compoundⅣ had significant effect on inhibiting the proliferation of Jurkat cellsat2mg/ml,0.5mg/ml0.125mg/ml and0.031mg/ml concentration(P<0.05);compoundⅥ had significant effect on inhibiting the proliferation of Jurkat cellsat1.92mg/ml,0.96mg/ml and0.48mg/ml concentration(P<0.05),respectively.Conclusion1. Enriched the non-alkaloid fraction by Macroporous Adsorption Resin andCation Exchange Resin.we got46.7g.2.6monomeric compounds were isolated.An ester compound(CompoundⅠ);two acids(CompoundⅡ,CompoundⅢ);a flavonoids(CompoundⅣ);a steroidalcompound(CompoundⅤ);a aldehydes(CompoundⅥ).The compoundⅠ,Ⅱ,Ⅲ,Ⅵwerefirst isolated from Aconitum flavum Hand.-Mazz..3.Non-alkaloids active fraction of Aconitum flavum Hand.-Mazz.,compoundⅠ,compoundⅡ, compound Ⅲ and compound Ⅵ inhibited the proliferation ofJurkat cells.
