PurposeTo study the effects of ?-Cymorphin-7 on the intestinal tissue morphology,intestinal epithelial lymphocytes,intestinal mucosal immune cytokines and intestinal mucosal oxidative stress in natural aging,preliminary study of P-Cytomorphin-7 affecting the intestine The molecular mechanism of mucosal immunity provides scientific information for the further development of ?-Cytomorphin-7.MethodsAfter 48 weeks of adaptive feeding of 48 male Kunming mice and 12 young male Kunming mice,44 elderly mice were randomly divided into low,medium and high dose groups and elderly model group,with 11 rats in each group;Only young mice were set as a youth control group.The dry powder ?-CM-7 was formulated into low,medium and high concentrations of 0.2×10-6 mol/L,1×10-6 mol/L and 5×10-6 mol/L in 0.9%physiological saline,respectively 0.5 ml of ?-CM-7 solution was administered per day,and the young control group and the elderly model group were intragastrically administered with an equal volume of 0.9%physiological saline.Weighed once a week,and the cervical vertebrae were sacrificed after 30 days of continuous gavage.The duodenum,jejunum and ileum segments of mice were taken.The small intestine VH,CD,V/C,mucosal thickness and intestinal epithelial lymphocytes were determined by HE staining.The jejunum mucosa of mice was used to measure IL-2,TNF-a,SIgA,SOD,MDA,CAT content,and the relative expression levels of NF-?B,TRAF6 and TLR4 genes were determined by real-time fluorescent quantitative PCR.Results1.Effect of ?-Cytomorphin-7 on intestinal tissue morphology and lymphocytes in aging miceCompared with the young group,the number of epithelial lymphocytes in the duodenum segment decreased(P<0.01),intestinal mucosal thickness increased(P<0.05),and V/C value decreased significantly(P<0.05);The number of lymphocytes in the jejunal segment epithelium decreased(P<0.05),V/C decreased(P<0.01);Mucosal thickness,villus length,V/C value of ileum segment decreased(P<0.01),and crypt depth increased(P<0.01).Compared with the model group,the dose group could increase the number of duodenal epithelial lymphocytes(P<0.01),the number of jejunal epithelial lymphocytes(P<0.05),and the high dose group could increase the number of ileal epithelial lymphocytes(P<0.05).The low dose group can significantly increase the length of duodenum and jejunum villi in mice(P<0.05),and increase the V/C value of three intestinal segments(P<0.01);The middle dose group can significantly increase the thickness of ileum mucosa(P<0.01),increase the length of duodenum and ileum(P<0.05),decrease the depth of ileum crypt(P<0.05),and increase the V/C value of duodenum(P<0.05),V/C values of jejunum and ileum(P<0.01);High dose group can increase the length of ileum intestine and the V/C value of jejunum and ileum(P<0.01).2.Effect of ?-Cytomorphin-7 on intestinal mucosal immune cytokines in aging miceThe TNF-a level in the elderly model group was higher than that in the young control group(P<0.01).Compared with the elderly model group,the low-and medium-dose groups significantly reduced the TNF-a content(P<0.05).The low-dose group significantly increased the IL-2 content in the senile small intestinal mucosa(P<0.05).The SIgA content in the low and middle dose groups decreased slightly(P>0.05),and the SIgA content in the high dose group was slightly higher than that in the old model group(P>0.05).3.Effect of(3-Cytomorphin-7 on body mass and intestinal mucosal oxidative stress in aging miceAt the end of the intervention,the weight of the dose group and the elderly model group decreased slightly compared with the initial weight.The weight of the young control group at the end of the intervention was slightly higher than the initial weight,But the difference is not statistically significant(P>0.05)The activity of SOD and CAT in the intestinal mucosa of the old model group was significantly lower than that of the young control group(P<0.01),and the MDA content was significantly higher than that of the young control group(P<0.01).Compared with the elderly model group,the low-and medium-dose groups significantly increased SOD in the small intestinal mucosa(P<0.01)and decreased MDA content(P<0.05).The low-dose group significantly increased CAT activity(P<0.01).High dose group can significantly reduce MDA content(P<0.01).4.Effect of ?-Cytomorphin-7 on TLR4,TRAF6 and NF-?B in intestinal mucosa of aging miceThe relative expression of TLR4,TRAF6 and NF-?B in the intestinal mucosa of the elderly model group was lower than that of the young control group,and the difference of TRAF6 and NF-?B was significant(P<0.05 or P<0.01).Compared with the model group,the low dose group could up-regulate the relative expression of TLR4 and TRAF6(P<0.01)and the relative expression of NF-?B(P<0.05).The medium dose could up-regulate the relative expression of TLR4 and NF-?B(P<0.01)and relative expression of TRAF6(P<0.05)ConsultsThe aging of the body leads to a change in the structure of the small intestine,a decrease in the content of antioxidant enzymes,an oxidative stress in the intestinal mucosa,an increase in the concentration of pro-inflammatory cytokines in the small intestinal mucosa,a decrease in the secretion of immune factors,and an inhibition of the relevant factors in the pathway of immunity.Activation reduces the function of the intestinal mucosal immune system.Ingestion of ?-CM-7 can alleviate the damage of aging on the structure of small intestine,stimulate the differentiation of intestinal epithelial lymphocytes,significantly increase the activity of antioxidant enzymes in small intestinal mucosa,reduce cell damage caused by peroxidation,and improve intestinal mucosal immune factors.Activity,reduce the content of pro-inflammatory factors in the small intestinal mucosa,improve the activation of immune-related genes,thereby enhancing the intestinal mucosal immune function.It is suggested that ?-CM-7 has a certain protective effect on the senile intestinal mucosal immune system.
