Objective: To observe the effect of electroacupuncture,combing with swimming on bone morphology and OPG expression in ovariectomized osteoporosis rats,and to explore the difference and possible mechanism of electroacupuncture and swimming exercise or not on osteoporosis in rats,The experimental basis for the treatment of osteoporosis.Methods: Sixty 6-8 months SPF SD female rats aged were numbered1-60.Twelve rats were randomly selected as sham-operated group(periovarian adipose tissue was removed)and the remaining 48 rats were removed to establish models.Bone mineral density was measured after 12 weeks of normal feeding.The rats were divided into model group,EA group,exercise group and EA exercise group according to body weight.There was no significant difference in body weight between groups.Acupoints of EA:group A(Mingmen,bilateral Shenshu and bilateral Pishu);the other group B(Zusanli and Guanyuan points on both sides);acupoints of group A and B were alternately carried out once a day,with 15 minutes of electrification each time and 10 minutes of needle retention.Swimming program was adopted for 45 minutes once a day.Six days of treatment is a course of treatment,one day of rest between courses,a total of eight courses of treatment.Routine feeding during treatment.At the end of the course of treatment,the body weight,bone mineral density,morphology of femoral bone tissue,expression of OPG protein and expression of OPG gene in bone marrow were observed.Results:1.Changes of body mass in rats: Before treatment,the weight of ovariectomized rats was higher than that of sham-operated rats(P<0.05),and the difference was statistically significant.After treatment,the body mass of rats in electroacupuncture group,exercise group and needle transport group was lower than that in model group(P<0.05),and the difference was statistically significant.The body mass of electroacupuncturegroup,exercise group and needle transport group decreased after treatment compared with before treatment(P<0.05),and the difference was statistically significant.2.Comparison of bone mineral density in rats: After treatment,the bone mineral density in model group was lower than that in sham-operated group(P<0.05),and the difference was statistically significant.Compared with the model group,the bone mineral density of the electroacupuncture group and the needle transport group was higher(P<0.05),and the difference was statistically significant.The bone mineral density of the needle transport group was higher than that of the electroacupuncture group(P<0.05),and the difference was statistically significant.3.Histological changes of femoral bone in rats: Sham-operated group had more complete bone structure,dense distribution and tighter arrangement of bone trabeculae.Compared with sham-operated group,the bone trabeculae in model group were obviously sparse,rough and discontinuous.The number of bone trabeculae in exercise group,electroacupuncture group and needle transport group was more than that in model group and the continuity was still acceptable.4.The expression of OPG protein and bone marrow OPG gene in femoral bone tissue of rats: After treatment,the expression of OPG in model group was lower than that in sham operation group(P<0.05),and the difference was statistically significant.The expression of OPG in EA group and needle transport group was higher than that in model group(P<0.05),and the difference was statistically significant.The expression of OPG in the needle transport group was higher than that in the electroacupuncture group(P<0.05),and the difference was statistically significant.Conclusions:1.Electroacupuncture combined with swimming exercise alone or in combination can reduce the body weight of ovariectomized rats with osteoporosis,and there is no significant difference in the degree of reduction.2.Electroacupuncture and swimming alone or in combination have positive effects on bone remodeling.Electroacupuncture and electro acupuncture combined with swimming exercise can improve bone tissue morphology,bone mineral density and OPG expression in ovariecto mized rats with osteoporosis.Compared with electroacupuncture alone,electroacupuncture combined with swimming exercise has synergistic effect.3.Swimming program has the tendency of improving bone histomorphology,increasing bone mineral density and promoting OPG expression,but its effect is not significant.
