Objective:to study the therapeutic mechanism of ningxin anshen recipe on generalized anxiety disorder(gad)by treating anxiety rats with uncertain empty bottle stress model and to initially explore its possible therapeutic targets.Methods:60 healthy Wistar male rats were randomly divided into blank group,model group,western medicine group,low dose Chinese medicine group,medium dose Chinese medicine group and high dose Chinese medicine group after 7 days of pre-culture.Among them,the blank group was given free diet,the model group was given normal saline gavage,the western medicine group was given lorazepam 0.05mg/(kg d),the middle dose group was given ningxinanshen prescription 11.625g/(kg d),the low dose group was 1/2,and the high dose group was twice as much.Behavioral tests(elevated cross maze,light and dark box)were performed after 14 days of stress.Blood and brain tissues(hippocampus,amygdala and prefrontal lobe)were collected after the experiment,and the contents of CCK,5-ht,GABA,NE and other indicators were detected by ELISA.Results:1.Behavioral experiment:compared with the blank group,The Times of entering the bright box,the percentage of journey and the percentage of time in the model group were reduced(P<0.05).Compared with the model group,the number of patients in the medium dose group increased(P<0.05),and the number of patients in the high dose group and the western medicine group increased significantly(P<0.01).Compared with the model group,the ratio of the distance to the open box increased in the high-dose Chinese medicine group and the western medicine group(P<0.05).Compared with the blank group,the percentage of the model group entering the open-arm maze was reduced(P<0.05).Compared with the blank group,the percentage of entering opening time in the model group was significantly reduced(P<0.01).Compared with the model group,the percentage of opening time in the high-dose group increased(P<0.05).There was no significant difference in the percentage of opening times between each group(P>0.05).2.Serum test results:compared with the blank group,CCK content in the model group was increased(P<0.05).Compared with the model group,CCK content in the western medicine group and the medium dose group decreased(P<0.05),while that in the high dose group decreased significantly(P<0.01).GABA content in the model group was significantly lower than that in the blank group(P<0.01).Compared with the model group,the content of GABA increased in the western medicine group(P<0.05),and the content of GABA increased significantly in the medium dose group and high dose group(P<0.01).NE content in the model group was significantly lower than that in the blank group(P<0.01).Compared with the model group,there was no significant difference among the treatment groups(P>0.05).5-ht content in the model group was significantly lower than that in the blank group(P<0.01).Compared with the model group,the content of 5-ht increased in the western medicine group,the medium dose group and the high dose group(P<0.05).3.Brain tissue detection results:CCK,(1)in the hippocampus,compared with the blank group,the CCK concentration in the model group decreased(P<0.01);Compared with the model group,CCK concentration increased in the medium dose group and the high dose group(P>0.05).(2)in the prefrontal lobe,CCK concentration in the model group was significantly lower than that in the blank group(P<0.01),Compared with the model group,the content of CCK increased in the western medicine group and the medium dose group(P<0.05),and the content of CCK increased significantly in the high dose group(P<0.01).(3)in the amygdala,compared with the blank group,the CCK content in the model group was significantly reduced(P<0.01).Compared with the model group,the CCK content in the medium dose group and the high dose group was significantly increased(P<0.01).GABA,(1)in the hippocampus,compared with the blank group,the content of GABA in the model group was significantly reduced(P<0.01).Compared with the model group,GABA content increased in the low-dose group(P<0.05).Compared with the model group,the contents of GABA in the western medicine group,the medium dose group and the high dose group were significantly increased(P<0.01).(2)in the prefrontal lobe,the content of GABA in the model group was significantly lower than that in the blank group(P<0.01).Compared with the model group,the content of GABA in the western medicine group and the medium dose group increased(P<0.05),while that in the high dose group increased significantly(P<0.01).(3)in the amygdala,compared with the blank group,the content of GABA in the model group was significantly lower(P<0.01).Compared with the model group,the contents of GABA in the western medicine group and the traditional Chinese medicine group were significantly increased(P<0.01).NE,(1)in the hippocampus,compared with the blank group,the content of NE in the model group was significantly reduced(P<0.05).Compared with the model group,the content of NE in the medium dose group increased(P<0.05),and that in the western medicine group and the high dose group increased significantly(P<0.01).(2)in the prefrontal lobe,compared with the blank group,the content of NE in the model group was significantly reduced(P<0.01).Compared with the model group,the content of NE in medium dose group and high dose group increased significantly(P<0.01).(3)compared with the blank group,the content of NE in the model group was significantly lower(P<0.01).Compared with the model group,the content of NE was significantly increased in the western medicine group,the medium dose group and the high dose group(P<0.01).5-ht,(1)in the hippocampus,compared with the blank group,the content of 5-ht in the model group was reduced(P<0.05).Compared with the model group,the content of 5-ht increased in the western medicine group and the medium dose group(P<0.05),and the content of 5-ht increased significantly in the high dose group(P<0.01).(2)in the prefrontal lobe,compared with the blank group,the content of 5-ht in the model group was significantly reduced(P<0.01).Compared with the model group,there was no significant difference among the treatment groups(P>0.05).(3)in the amygdala,compared with the blank group,the content of 5-ht in the model group was significantly reduced(P<0.01).Compared with the model group,the contents of 5-ht were significantly increased in the western medicine group,the medium dose group and the high dose group(P<0.01).Conclusion:(1)Ningxin anshen granules can regulate the concentration of GABA in brain tissue and blood to play an anti-anxiety role.The effect of regulating the concentration of GABA in brain tissue is similar to lorazepam,but lorazepam cannot regulate the concentration of GABA in blood.Ningxin anshen granules can act on the CCK system and regulate the balance of CCK concentration in serum and brain tissue to play an anti-anxiety role,which is different from lorazepam.(2)both medium and high doses of ningxin anshen granules have anti-anxiety effects,and the effect is greater than or equal to one tablet of lorazepam.(3)the dose of ningxinanshen granules in the high-dose group was twice that of the medium-dose group,but the curative effect between the two groups showed no multiple relationship.Ziziphus ziziphus seed and acanthopanax acanthopanax were expensive drugs,and the high-dose group would increase the economic burden of patients,so the medium-dose group was more suitable for clinical use.
