Objective:To study the effect of Weitong xiaopi prescription on the changes of the general conditions,small intestine advancing rate,residual rate in the stomach,and the CGRP in gastric antrum tissue and hypothalamus,during and after drug treatment in rats with functional dyspepsia of liver depression and spleen deficiency type.And the effect of CGRP-mRNA expression reveals the mechanism of action of stomach carbuncle elimination in the treatment of functional dyspepsia,thus providing scientific and objective experimental data and theoretical basis for clinical treatment of functional dyspepsia.Methods:1.Modeling:The male SD rats were randomly divided into 4 groups according to the blank control group,the model control group,the Weitongxiaopi prescription group and the mosapride group,a total of 48 rats with 12 in each group.In addition to the blank control group,the rest of the groups were used Guo clip tail stimulation combined with Zhang irregular feeding method to create rat model of liver-qi depression and spleen deficiency.2.After successful establishing model,the general condition of rats in each group was recorded,and the blank control group and the model control group were not treated.Weitongxiaopi prescription group and Mosapride group were given granule of Weitongxiaopi prescription and Mosapride tablets diluted solution on the basis of the model respectively,according to the body surface area conversion method between human and rat,twice a day,continuous administration 21 days.After the treatment,the general condition score of each group was observed.The rats in each group were fasted for 24 hours,and then the rats in each group were subjected to abdominal anesthesia in turn.The relevant parts of the rats were removed and then the residual rate and the small intestine propulsion The expressions of CGRP and CGRP-mRNA in gastric antrum and hypothalamus of rats in each group were detected by Western blotting and real-time fluorescence quantitative PCR respectively.4.SPSS20.0 software was used to process the data and expressed as(mean ± standard)deviation.Single factor analysis of variance(ANOVA)was used to satisfy the normality.ANOVA LSD test was used to compare the variance among groups.If variance was not good,variance analysis t test.If P<0.05 there are differences between the two groups,and the difference was statistically significant.Result:1.Compared with the blank control group,the control group of the model decreased in general state,such as luster,activity,diet and weight change.Compared with the control group,the Weitongxiaopi prescription group and Mosapride group were improved(P<0.05),and the Weitongxiaopi prescription group was better than the Mosapride group(P<0.05).2.Compared with the blank control group,the intestinal propulsion rate of the model control group decreased(P<0.05);compared with the model control group,the rate of small bowel propulsion was increased in the Weitongxiaopi prescription group and Mosapride group(P<0.05),and the Weitongxiaopi prescription group was better than the Mosapride group.3.Compared with the blank control group,the residual rate in the stomach of the model control group increased(P<0.05);compared with the model control group,the residual rate in the stomach decreased in both the Weitongxiaopi prescription group and Mosapride group(P<0.05),and the Weitongxiaopi prescription group was better than the Mosapride group.4.Compared with the control group,the expression of CGRP protein and CGRP mRNA in the gastric antrum and hypothalamus increased in the model control group(P<0.05).Compared with the model control group,the Weitongxiaopi prescription group and Mosapride group The expressions of CGRP protein and CGRP mRNA in gastric antrum and hypothalamus decreased(P<0.05),and the Weitongxiaopi prescription group was better than the Mosapride group.Conclusion:1.Using Guo's clip tail stimulation combined with Zhang's irregular feeding method can successfully replicate liver depression and spleen deficiency FD rat model.2.Weitongxiaopi prescription can significantly improve the general state of FD rats,food intake,weight gain,intestinal propulsion rate,and residual rate in the stomach,which has a positive effect on gastrointestinal motility.3.Weitongxiaopi prescription can effectively regulate CGRP content and gene expression in gastric antrum and hypothalamus of FD rats with liver depression and spleen deficiency.This experiment proved thatWeitongxiaopi prescription can improve gastrointestinal motility disorders,reduce the gastrointestinal tract sensitivity,regulate the level of brain-gut peptide secretion,and have a significant effect on the treatment of FD.4.The effect of Weitongxiaopi prescription on functional dyspepsia may be superior to Mosapride...
