Objective:1.The purpose of this paper was to probe into the realationship between HPRL and living factors,which including dietary habits,emotions and sleep quality,with epidemiological survey.2.Take Liver and Spleen as the core then to investigate effect of Jiawei-xiaoyaosan on dopamine receptor P38MAPK signal transduction pathway in hyperprolactinemia(HPRL)rats.Methods:1.Divided volunteers into control group and HPRL group.Investigated these volunteers who met the inclusion criteria with uniform questionnaires randomly,and then SPSS 18.0 software was applied for statistical treatment.2.We set up animal models of HPRL by using pituitary transplantation,then divided HPRL rats randomly into blank control group,model group,bromocriptine group,antagonist of D1 receptor groups and Chinese medicine groups.Antagonist of D1 receptor groups included SCH23390 group,SCH23390 combining Chinese medicine middle dose group and SCH23390 combining bromocriptine group.Chinese medicine groups included high dose group,middle dose group and low dose group.Fed these rats 30 days accordingly,then determine the relative expression quantity of LPS,PAK,P38MAPK in the hypothalamus of each group by immunohistochemical staining,western blotting and RT-PCR method.Results:1.In aspect of diet,11 cases of having meals not in time in control group(7.3%),25cases in HPRL group(16.7%),38 cases of preferring fry in control group(25.3%),55 cases in HPRL group(36.7%),34 cases of preferring meaty diet in control group(22.7%),61 cases in HPRL group(40.7%),22 cases of having snacks in control group(14.7%),39 cases in HPRL group(26%),9 cases of smoking in control group(6%),16 cases in HPRL group(10.7%)including 11 cases of smoking 1 to 5 cigarettes per week(7.3%),4 cases of smoking 6 to 10 cigarettes per week(2.7%),1 cases of smoking 11 or more per week(0.7%),64 cases of drinking in control group(42.7%),103 cases in HPRL group(68.7%)including 39 cases of drinking 1 to 2 times per week(26%),47 cases of drinking 3 to 5 times per week(31.3%),17 cases of drinking 6 or more times per week(11.3%).In aspect of anxiety,the average score of control group was 37.46,and 17 cases of having anxiety(11.3%),while the average score of HPRL group was 55.49,and 98 cases of having anxiety(65.3%),including 31 cases of mild anxiety(20.7%),49 cases of medium anxiety(32.7%),18 cases of severe anxiety(12%).In aspect of depression,the average score of control group was 38.13,and 21 cases of having depression(14%),while the average score of HPRL group was 51.51,and 83 cases of having depression(55.3%),including 43 cases of mild depression(28.7%),30 cases of medium depression(20%),10 cases of severe depression(6.7%).In aspect of sleep,the average score of control group was 8.57 and 47 cases of having high-quality(31.3%),17 cases of having inferior-quality(11.3%),while the average scores of HPRL group was 11.01,and 26 cases of having high-quality(17.3%),23 cases of having inferior-quality(15.3%).2.The animal experiment showed that Jiawei-Xiaoyaosan could effect the P38MAPK signal transduction pathway of HPRL rats to some degree.Compared with the blank control group,the quantity of LPS,PAK,P38MAPK in hypothalamus of model group rose significantly.Applying the antagonist of D1 receptor,compared with the model group,the quantity of LPS,PAK,P38MAPK in SCH23390 group reduced significantly.Compared with the SCH23390 group,SCH23390 combining Chinese medicine middle dose group could reduce the quantity of LPS and PAK slightly,but had obvious inhibitional effect on P38MAPK.Without any antagonist of receptor,compared with the model group,the quantity of LPS,PAK,P38MAPK in each Chinese medicine group rose significantly,especially in Chinese medicine high dose group.Bromocriptine had less effect anbout LPS,PAK,P38MAPK than Chinese medicine.Conclusion:1.As most of patients with HPRL have unhealthy habits,nagetive moods and poor sleep,we can concluded that eating,emotion and sleeping may impact secretaion and regulatory functions of hypothalamus on PRL.2.The effect of Jiawei-xiaoyaosan of dopamine receptor P3 8-mitogen-activated protein kinase(P38MAPK)signal transduction pathway is inhibitive.This prescription can restrain D1R,obstruct LPS combine with receptor,then reduce the expresstion of quantity in some related intermediates by depressing thePAK—P38MAPK reaction chain.Chinese medicine groups have a concentration-response relationship.Chinese medicine may restrain DIR to promote D2R combine with DA,so the level of PRL will be reduced.
