Exploring The Action Mechanism Of Qutanhuoxue Granules Regulating AQP7 In NAFLD Model Rats Adipose Tissue Based On The P38MAPK Pathway

Objective: To observe the expression of AQP7 and P38 MAPK in NAFLD rats adipose tissue after being treated with Qutanhuoxue granules and explore the mechanism for treating with Qutanhuoxue granules in NAFLD rats.Methods: Choosing 40 clean grade male SD rats of body weight in 180~220g with normal diet adaptive feeding one week,than all rats were randomly divided into normal group(n=6)and model group(n=34);Normal control group was fed with common diet,model group was fed with high fat diet in order to induce NAFLD rats model.One or two rat was randomly selected to kill every 2 weeks begine with tenth weeks,then confirming whether NAFLD model was successful or not by the liver pathological staining,observing the pathological changes;Confirmed the success of model,then the remaining model rats were randomly divided into model group(n=6),Chinese medicine high dose group(n=6)?middle dose group(n=6)?low dose group(n=6),P38 MAPK inhibitor group(n=6);The Chinese medicine groups were fed with qutanhuoxue granules and inhibitor group was given with SB203580 intraperitoneal injection of P38 MAPK inhibitor(SB203580)for intervention;For 4 weeks,then all rats were killed,observing the changes of liver HE staining and oil redO staining with light microscopy,determinating serum TG,TC,AST,ALT,liver TG,FFA,liver index and the expression of AQP7 and P38 MAPK gene and protein in adipose tissue.Results:1.Liver pathological HE and oil red O staining showed: Compared with normal group,a increase number of intracellular lipid droplets and hepatic steatosis is obvious in model group rats,the difference was statistically significant(P<0.05);Compared with the model group,the liver intracellular lipid droplets of Chinese medicine group and inhibitor group rats decreased,with showing the degree of liver steatosis reduced significantly(P<0.05);2.Serum biochemical indicator showed:Compared with normal group,the model group rats serum TG,TC,AST and ALT increased significantly;Compared with model group,the Chinese medicine groups and inhibitor group rats serum TG,TC,AST and ALT decreased significantly(P<0.05);3.Liver TG,FFA and liver index showed: Compared with normal group,the model group rats liver TG,FFA and liver index increased significantly;Compared with model group,the Chinese medicine groups and inhibitor group rats liver TG,FFA and liver index decreased significantly(P<0.05);4.Realtime-PCR results showed: Compared with normal group,the model group rats showed a significantly decrease in the expression of AQP7 m RNA and a increase of P38 MAPK m RNA in adipose tissue(P<0.05);Compared with model group,the Chinese medicine groups and inhibitor group ratsshowed a significantly increase in the expression of AQP7 m RNA,which high dose Chinese medicine group had the best effect,and a decrease of P38 MAPK m RNA,which the inhibitor group had the most obvious effect(P<0.05);Chinese medicine low and middle dose group have no significant difference in the expression of AQP7 and P38 MAPK m RNA(P>0.05);5.Detection of proteins results showed: Compared with normal group,the model group rats showed a significantly decrease in the expression of AQP7 protein and a increase of p-P38 MAPK protein in adipose tissue(P<0.05);Compared with model group,the Chinese medicine groups and inhibitor group rats showed a significantly increase in the expression of AQP7 protein,which high dose Chinese medicine group had the best effect,and a decrease of p-P38 MAPK protein,which the inhibitor group had the most obvious effect,and the effect of high dose Chinese medicine group taken second place(P<0.05);Traditional Chinese medicine low and middle dose group had no significant difference in the expression of AQP7 and P38 MAPK protein(P>0.05);There was no significant difference in total P38 MAPK protein in adipose tissue of each group rats(P>0.05).Conclusion: 1.Qutanhuoxue granules can improve the pathological structure of NAFLD rats,reduce the degree of liver steatosis and liver inflammation;2.Qutanhuoxue granules can reduce NAFLD rat liver index,serum TG,TC,AST,ALT and liver TG,FFA;3.Qutanhuoxue granules probable caninhibit the activation of P38 MAPK and up-regulate the expression of AQP7 in adipose tissue,increase plasma glycerol metabolism,reduce glycerol and FFA intaked into liver,hereby reducing liver fat accumulation,and reducing liver lipid fatty degeneration degree;4.The activation of the P38 MAPK pathway may be involved in inhibiting the expression of AQP7 in adipose tissue,which can regulate the metabolism of glycerol and fat,thus indirectly improving NAFLD.
