ObjectiveTo select miRNAs of chronic superficial gastritis patients with Qi deficiency constitutional health and spleen deficiency syndrome to compare with those of healthy subjects with moderate constitutional health to explore the miRNA involved in the development of Qi deficiency constitution-leukocyte miRNA markers in patients with chronic superficial gastritis.MethodsSubjects were enrolled in the Hepatobiliary and Gastroenterological Outpatient Department of Dongfang Hospital of Beijing University of Chinese Medicine between September 2016 and July 2017.After screening and inclusion criteria were included in the screening criteria,3 healthy and moderately healthy subjects 2 cases of people,3 cases of chronic superficial gastritis patients with spleen deficiency syndrome were studied,there was no significant difference in gender and age between groups.The leukocyte of healthy people with moderate constitutional health,healthy people with deficiency of constitution and patients with chronic superficial gastritis of spleen deficiency syndrome were respectively isolated.After total RNA was extracted and sequenced,the miRNA expression profiles were analyzed.According to the miRNA target genes in the database The GO and KEGG analyzes predict the biological processes involved in the differential expression of miRNAs.ResultsHCL clustering analysis of serum miRNA expression profiles of normal healthy people,healthy people with deficiency of qi and chronic superficial gastritis with spleen deficiency syndrome showed that the miRNA expression profile could distinguish three groups well,among them,The group with chronic constitutional gastritis with spleen deficiency syndrome was relatively far away from the former two groups and was in line with the experimental expectation.There were 31 miRNAs(P<0.05)differentially expressed between healthy volunteers and healthy volunteers,of which 13 were up-regulated(fold change>1.5)and 8 were down-regulated(0.5 fold)Pathway analysis showed that the GO biological process analysis was mainly associated with extracellular matrix,cell junctions,signal transduction,substance and energy metabolism processes,KEGG analysis was mainly associated with extracellular matrix,cell junctions and signal transduction pathways,Biological analysis of the results are similar,the main signaling pathways involved include Hppo signaling pathway,TGF-? signaling pathway,FOXO signaling pathway and p53 signaling pathway;spleen deficiency chronic superficial gastritis patients compared with normal physical health 38 miRNAs were differentially expressed(P<0.05),of which 25 were up-regulated(fold change>1.5)and 13 were down-regulated(0.5 fold).The analysis of GO and KEGG pathway showed that the biological processes of GO were mainly related to Metabolism,gene expression and other processes related to KEGG analysis and extracellular matrix,substance synthesis and metabolism,signal transduction pathways,the main signaling pathways involved include Hi ppo signaling pathway and p53 signaling pathway.Among them,the levels of miR-145-5p and miR-146a-3p tended to increase with the order of patients with chronic superficial gastritis of healthy people with constitutional deficiency-qi deficiency and healthy people-spleen deficiency syndrome.The GO biology process analysis and KEGG pathway analysis of these two miRNAs showed that the biological processes involved in the two miRNAs are mainly related to the synthesis and metabolism of materials,and the common pathway is the Hippo signaling pathway.ConclusionThe up-regulation of miR-145-5p and miR-146a-3p can be observed in serum of patients with deficiency of constitutional health and chronic superficial gastritis of spleen deficiency syndrome,and with the development of the disease,there may be an upward trend and may be used as the constitution and syndrome of qi deficiency Candidate biomarkers.There may be some connection between the Hippo signaling pathway and the pathogenesis of chronic superficial gastritis in spleen-deficiency syndrome,which is worth further exploration.
