Objective:To observe the effects of the spontaneous activity circadian rhythm of insomnia SD rats based on the treating with acupuncture in Shenmai(BL62)and Zhaohai(KI6).According to the change of contents of the MT,5-HT,NE,expression level of the MT1mRNA inside the SCN to investigate the internal mechanism of acupuncture adjusting the sleep-wake cycle based on Qiao-meridian theory,to provide theoretical basis for clinical treatment of insomnia.Methods:I.When the 30 SD rats act with the light from the darkness steadily,then divided them into three group randomly,that was blank,model and acupuncture,with animal recording and analyzing system to observe spontaneous activity circadian rhythm.2.Use intraperitoneal injection of PCPA suspension to copy insomnia SD rats model,and observe the characteristic of spontaneous activity circadian rhythm in SD rat model of insomnia,measured by the changes of daily spontaneous activity.3.After the success of copying model of insomnia,acupuncture was applied to Shenmai(BL62),and Zhaohai(KI6)for 20 minutes per day,and lasted for 7 days during 8:00-10:00am.Then observe the changes of them.4.The contents of MT were detected by HPLC method,the contents of MT1mRNA were detected by RT-PCR method,the contents of 5-HT?NE were detected by ELISA method.Results:1.In L:D(12:12)conditions,the SD rats were show the stable spontaneous activity circadian rhythm,There was no statistically significant difference between the rest period activity of groups(p>0.05).But the disorder of spontaneous activity circadian rhythm was expressed by the model of insomnia.The change of insomnia model was the increase of daily spontaneous activity(p<0.01).2.After Acupuncture treatment,compared with model group,the reduction of daily spontaneous activity could be observed in acupuncture group(p<0.01).3.After injection of PCPA,compared with blank group,the content of MT,5-HT were decreased(p<0.01),the expression of MT1mRNA was decreased(p<0.05),but the content of NE was increased in model group(P<0.01).4.After Acupuncture treatment,compared with model group,the content of MT,5-HT were increased(p<0.01,p<0.05),the expression of MT1mRNA was increased(p<0.05),but the content of NE was decreased in acupuncture group(P<0.05).Conclusions:1.Insomnia SD rats spontaneous circadian rhythm disorders,Acupuncture treatment could significantly reduce the amount of daily spontaneous activity to promote sleep.2.The content of MT,5-HT were decreased,the expression of MT1mRNA was decreased,but the content of NE was increased in insomnia SD rats.The mechanism of why acupuncture Qiao-meridian could treat insomnia perhaps is related with increasing the content of MT and the expression of MT1mRNA,decreasing the content of NE and IL-1? to promote the release of 5-HT,inhibit the release of the NE,recovering nervous-endocrine system,Regulating sleep-wake cycle.
