Objective:To explore the effects of electric-acupuncture on the wake-sleep balance and the DMH-VLPO and DMH-LHA feedback under advances of light/dark cycle, in order to uncover the mechanism of acupuncture adjusting the rhythm disturbance.Method:SD rats were firstly synchronized to normal light/dark cycle for a week, we duplicate the rhythm disorders by5cycles of serial8-hour advances LD cycle every2days. After that, using immunohistochemistry&image analysis system and ELIS A, to observe the effects of electric-acupuncturing both "Nei guan(PC6)" and "Zu sanli(ST36)" at ZT4on EEG and associated neurotransmitters in the brain of SD rats by immunohistochemistry.Results:l.Before advancing light/dark cycle, there’s no significant differences in the phase shift among the groups of the time of wake、TST、SWS-2and PS of animals.2.The time of TST、SWS-2and PS and the expression of GABA accepter in DMH and the GAL in VLPO was significantly decreased,and the expression of Orexin protein in LHA and the L-Glu accepter in DMH was significantly increased.3.After treatment at ZT4,the time of TST、SWS-2and PS and the expression of GABA accepter in DMH and the GAL in VLPO were significantly increased;the expression of Orexin protein in LHA and the L-Glu accepter in DMH was significantly decreased.Conclusions:As a non-light timing factor,the mechanism of Electric-acupuncture regulating rhythm resynchronization and the imbalance of wake-sleep may be as follows:1.Electric-acupuncture at ZT4can significantly reduce L-Glu accepter and Orexin concentrations, to maintain a normal circadian rhythm,to ensure the normal activity-rest rhythm.2.1t can work on the DMH-VLPO and DMH-LHA feedback by up-regulating the expression of GABA accepter within the DMH and the GAL in VLPO to inhibit the activity of neurons and maintain normal sleep-wake rhythm,and by inhibiting the expression of L-Glu accepter within the DMH and the Orexin in LHA to delay the time,thereby promoting the resychronization of circadian rhythms.
