The Expression Of MiRNA-194 In Breast Cancer Cells And Its Significance | Posted on:2019-09-28 | Degree:Master | Type:Thesis | Country:China | Candidate:G Q Dai | Full Text:PDF | GTID:2434330566489604 | Subject:General surgery | Abstract/Summary: | PDF Full Text Request | [Objective] To explore the effect of mi RNA-194 on the chemosensitivity of breast cancer cells and the relatonship between mi RNA-194 and the clinical response of docetaxel in breast cancer patients.[Methods] Real-time PCR was used to detect the expression of mi RNA-194 in breast cancer HCC1937,MCF-7 cells and human normal mammary gland cell line 578 Bst.The mi RNA-194 mimics ion was transfected.The effect of mi RNA-194 on the IC50 of docetaxel in breast cancer HCC1937 and MCF-7 cells was detected by MTT assay.Real-time PCR was used to detect the difference of mi RNA-194 expression in breast cancer tissues and adjacent mucosa.The relationship of mi RNA-194 and the clinical response of docetaxel in breast cancer patients was analysed by Spearman rank test.[Results] The expression level of Mi RNA-194 in HCC1937 and MCF-7 cells was significantly lower than that in Hs 578 Bst cells(P <0.05).After transfection,the level of mi R-194 in HCC1937-mi R-194 and MCF-7-mi R-194 cells were significantly higher than those in untransfected and transfected NC group.The IC50 values of HCC1937 and MCF-7 were(6.32 ± 0.82)?mol / L and(1.13 ± 0.21)?mol / L,which were significantly higher than those in the transfected group[IC50 HCC1937-mi R-194=(2.98 ± 0.67)? mol / L,IC50 MCF-7-mi R-194=(0.42 ± 0.10)? mol / L](P <0.05).In clinical tissue samples,the expression level of mi R-194 in the adjacent tissues was significantly higher than that in the tuomr tissues(P <0.05).The expression level of mi RNA-194 was found negatively-correlated with lymphatic metastasis and tumor TNM classification(p<0.05).High expression level of mi R-194 was found positively-correlated with high chemosensitivity(Spearman rank test rho = 0.594,p<0.05).[Conclusion]The mi RNA-194 is dysregulated in breast cancer cells HCC1937 and MCF-7,and its high expression can increase the killing effect of docetaxel on HCC1937 and MCF-7 cells.There was a significant difference between the expression level of mi RNA-194 in cancer tissues and adjacent tissues.The expression level of mi RNA-194 was found negatively-correlated with lymphatic metastasis and tumor TNM classificationand high expression level of mi RNA-194 was positively-correlated with high chemosensitivity the clinical response of docetaxel... | Keywords/Search Tags: | Breast cancer, miRNA-194, Docetaxel, Chemosensitivity | PDF Full Text Request | Related items |
