Swimming Exercise Improves The Heart Function Of Spontaneously Hypertensive Rats By Up-regulating The CTRP9-AMPK-eNOS-NO System

Research objective:Spontaneous hypertension often involves cardiac target organs,which seriously endangers the quality of life and life style of human.Therefore,Spontaneous hypertension and control of hypertension is now a hot research project.Long term continuous hypertension can not only change the structure of the heart,but also reduce the heart function and myocardial damage.Over the years,some experiments have shown that CTRP9 has played a positive role in protecting the myocardium.Moderate exercise can improve the structure and function of the heart.The purpose of this study was to observe whether exercise could affect the expression of CTRP9 in myocardium of hypertensive rats,and the expression of associated protein AMPK,eNOS,and adiponectin receptor 1 through aerobic exercise intervention,so as to explore whether the exercise would increase the CTRP9-AMPK-eNOS-NO system.put forward a theoretical basis and reference data for prevention and treatment of hypertension.Research methods:Four week old male WKY rats were 20 rats,25 SHR rats were(the accidental death of a),feeding a week were randomly assigned to it.WKY rats were randomly divided into 9 groups:quiet group(WKY group),exercise control group 11(group WE),SHR rats were randomly divided into hypertension quiet group 10(SHR group),hypertension exercise control group 14(SE group).The cage is kept in a cage with 4-5 cages per cage.The room temperature is around 23 degrees centigrade,and the water is free.Every 2-3 days is replaced.After buying,the rats first adapted to the environment for a week,adapted for a week,started from second weeks,and took a daily swimming exercise with increasing time every day,training 15min every day until training time increased to 60min per day.After the completion of the adaptive training,six weeks exercise program without weight negative swimming was implemented.During the training process,weight was weighed and recorded weekly.After sixweeks of exercise cycle,the hemodynamics of rats were measured after 24h.Hemodynamic analysis of heart rate(HR),left ventricular systolic pressure(LVSP),left ventricular end diastolic pressure(LVEDP),left ventricular pressure increased the maximum rate(+dP/dtmax),and left ventricular pressure decreased the maximum rate(-dp/dtmax)and other cardiac function indicators.In the later period,the animal tissues were observed by HE staining,the detection of Western blot,the detection of the Elisa kit and so on.Research results:(1)The body weight of rats and the heart weight to body weight:six weeks swimming after intervention,compared with the WKY quiet weight group rats,SHR group was significantly lower than WKY group,WE group obviously decreased;the ratio of heart weight to body weight index,with the WKY group quiet group rats compared to the increased WE group and SHR group data.The above results indicate that hypertension can slow down the growth rate of body weight,and swimming has an inhibitory effect on weight gain of WKY exercise group and SHR exercise group.(2)The analysis of the function of heart hemodynamic index:four groups of rats according to hemodynamic index data shown compared with WKY rats,WE rats LVSP slightly increased,LVEDP decreased,+dp/dtmax,-dp/dtmax value increased significantly,indicating that exercise can improve heart function in rats;compared with WKY rats of group SHR,LVEDP,LVSP + dp/dtmax index showed a downward trend,indicating that the heart function of hypertension.Compared with SHR group,the LVSP of SE group increased,LVEDP and dp/dtmax value decreased significantly,and the trend tended to WE rats.The above results showed that exercise could improve cardiac function in hypertensive disease.(3)HE staining of tissue morphology results showed:WKY control group rat cardiac cells of normal distribution,the shape distribution is regular,the nucleus was oval or round;WE group the myocardial distribution is regular,out of shape with direction consistency,compact arrangement,myocardial fiber integrity,no scar;myocardial cells were SHR group of rats is messy,myocardial fibers or broken,or nuclear fusion,out of order distribution;myocardial cells of rats in the SE group compared with SHR comparison,cell shape improvement,nucleus distribution.(4)The detection results of Western blot showed that compared with WKY group,the expression level of CTRP9,AdipioRl,AMPK and eNOS increased in different degrees in group WE,and the expression in SHR group decreased significantly.Compared with the SHR group,the expression of group SE increased.These results suggest that the expression of molecules can be suppressed in the state of hypertension,and long-term aerobic exercise can slow down this inhibitory effect.(5)Elisa kit test results showed:compared with the rats in the WKY group,WE group was CTRP9 higher than that of WKY group,the rats in group SHR content expression level is lower than the WKY group;compared with SHR rats,SE rats increased content;the hypertension to reduce the content of CTRP9,aerobic the movement can increase the content of CTRP9.(6)Nitric oxide(NO)kit.The test results show that:compared with the rats in the WKY group,NO SE group were higher than that of WKY group,SHR group than in the WKY group is low,and there are obvious differences;compared with SHR rats,the rats of group SE expression increased;this can indicate high blood pressure can make the content of NO decreased,the content of exercise can upregulate NO.Conclusion:(1)Impaired myocardial structure and decreased myocardial function in spontaneously hypertensive rats.The expression level of CTRP9 protein was decreased,and the expression level of AMPK protein and eNOS protein decreased.The content of NO is also reduced.(2)Long-term aerobic exercise can improve cardiac structure and function in spontaneously hypertensive rats,which may increase myocardial CTRP9 protein expression,AMPK protein and eNOS protein expression.It may be associated with exercise by raising the CTRP9-AMPK-eNOS-NO system to give play to the heart.
