Zhenwu Decoction Regulates The TOLL-like Receptor/NF-?B Signaling Pathway In The Kidney Tissue Of Rats With Heart And Kidney Yang Deficiency

Objective:The effect of Zhenwu Decoction on left ventricular ejection fraction(LVEF),short axis shortening rate(LVFS),left ventricular end systolic diameter(LVIDs),serum brain natriuretic peptide(BNP)in rat,and the expression of TLR2(TLR2),Toll like receptor 4(TLR4)and nuclear factor kappa B(NF-NF-B)protein in rat with syndrome of yang deficiency of heart and kidney were investigated.Study on the pathogenesis of cardio renal syndrome.Treatment of traditional Chinese medicine with traffic heart and kidney.To provide experimental data and theoretical basis for clinical application of Zhenwu Decoction in treating cardio renal syndrome.Materials and methods:There were 55 healthy SD rats weighing 200 to 250g,half male and half female.After feeding for 1 weeks,10 were selected randomly as normal group,10 as sham operation group,and 35 as operation group.The bilateral thyroid gland was stripped from the rats by operation.Intraperitoneal injection of 0.02%adriamycin 6ml per kg,two times a day for three weeks in ten days after the operation for rat model of syndrome of yang deficiency of heart and kidney.After six weeks of modeling,the LVEF and LVFS values of the rats were detected by echocardiography.The serum NT-proBNP value was detected by ELISA method to determine whether the model was successful.In the operation group,thirty rats survived.According to random number table,fifteen rats were randomly selected as Zhenwu Decoction group,and the other fifteen as model control.The equivalent dose of rats was converted into the dosage of rats according to the dosage of human body.The dosage of Zhenwu Decoction and equivalent dose of two hundred grams of rats were 0.756g per day.And gavage of the stomach every day for eight weeks.Result:1.After modeling,the LVEF and LVFS levels in the operation group were significantly lower than those in the normal group.The serum NT-proBNP level of the operation group was higher than that of the normal group and the sham operation group.The difference was statistically significant(P<0.05).It shows that the model is successful.2.After eight weeks of medication,the serum NT-proBNP concentration in model group was higher than that in normal group and sham operation group.The difference was statistically significant(P<0.05).It is indicated that Zhenwu Decoction can effectively improve cardiac function in rats with renal failure and heart failure.3.After eight weeks of medication,compared with the normal group and sham operation group,the LVEF and LVFS values of the model group were significantly lower than those of the normal group and the sham operation group,and the LVIDS value increased;Compared with the model group,the LVEF and LVFS values of Zhenwu Decoction group increased significantly,while the LVIDS value decreased.And the difference was statistically significant(P<0.05).It is indicated that Zhenwu decoction is effective in improving the ejection function of rats with renal failure and heart failure.4.Pathological changes of renal tissue by HE staining:In the model group,the size of the glomeruli was reduced and the fibrosis was formed,the balloon was inflammatory exudation,the capillary ball and the vascular lumen were pressed,and the proliferation of mesangial cells and capillary endothelial cells was compared with that of the normal group.Renal tubular epithelial cell exfoliative necrosis.In the sham operation group,the glomerulus was of moderate size,the balloon had no obvious inflammatory exudation,and there was no obvious epithelial cell loss and necrosis in the renal tubules.Zhenwu Decoction group:glomerular size is moderate,no obvious inflammatory exudation in the balloon,and a small amount of renal tubular epithelial cells edema.It indicates that Zhenwu Decoction can effectively alleviate renal tissue lesions.5.Expression of Toll like receptor 2(TLR2),Toll like receptor 4(TLR4)and nuclear factor kappa B(NF-kappa B)protein in renal tissue:Immunohistochemistry and real-time quantitative PCR were used to detect the expression of TLR2,TLR4 and NF-kappa B protein in kidney tissue.Compared with normal group,the expression of TLR2,TLR4 and NF-kappa B protein in kidney tissue of model group was significantly higher than that of normal group(P<0.01).It indicates that there is inflammatory reaction in the process of heart kidney yang deficiency.TLR2,TLR4,NF-kappa B protein pathway is activated,and injury of the physiological function of the two organs of the heart and kidney.After eight weeks of medication,the expression of TLR2,TLR4 and NF-kappa B protein in rat kidney tissues was significantly lower than that in the model group(P<0.01).It indicates that Zhenwu decoction has a significant effect on reducing the levels of TLR2,TLR4 and NF-kappa B protein in kidney tissue of heart kidney yang deficiency syndrome.Conclusion:Zhenwu Decoction can effectively improve the LVEF and LVFS values of the rats with renal failure and heart failure,and reduce the LVIDS value.The level of serum NT-proBNP in rats was significantly reduced.Reduce the expression of TLR2,TLR4 and NF-kappa B protein in kidney tissue.It can obviously improve the heart function of the heart kidney yang deficiency rats and slow down the pathological changes of the kidney.lt indicates that the TLR like receptor/NF-kappa B protein pathway is involved in the progression of CRS.It can slow down the progression of CRS by inhibiting the expression of TLR2 and TLR4/NF-kappa B,and protect the two organs of the heart and kidney.
