Objective: Salvia chinensia Benth is a traditional Chinese medicine(TCM),it can promote the blood flow,remove the heat and detoxification,reduce swelling and pain,commonly used to treat infectious diseases and gynecological diseases.Pharmacological experiments have shown that,Salvia chinensia Benth has a good anti-cancer effect on a variety of malignant tumors.In this experiment,we prepared Salvia chinensia Benth drug-containing serum and used it to treat breast cancer cell MCF-7.We observed the effect of Salvia chinensia Benth on the proliferation and apoptosis of breast cancer cells,and the influence of microRNA-21 and Bcl-2 expression.To explore the anti-cancer mechanism of Salvia chinensia Benth,and provide a new evidence and basis to clinical drug use for the treatment of breast cancer.Methods: In this study,Salvia chinensia Benth is the research object.32 SPF female SD rats were screened and divided into A,B,C and D groups randomly.They were Salvia chinensia Benth group,traditional Chinese medicine compound group,positive control group(cyclophosphamide)and blank control group(saline)respectively.The rats in the four groups were given gavage daily and the blood was collected via abdominal aorta on the 7th day.Serum was separated,and drug-containing serum was obtained.Breast cancer cell MCF-7was treated with medicated serum.the proliferation inhibition rate of breast cancer cells was observed by cck8 experiment.The apoptosis of breast cancer cells was detected by flow cytometry.The expression levels of MicroRNA-21 and Bcl-2 were detected by real-time PCR and Western Blot respectively,to explore the molecular mechanism of anti-breast cancer effect of Salvia chinensia Benth.Result : 1.The effect of Salvia chinensia Benth on Proliferation and Apoptosis of Breast Cancer Cell MCF-7In CCK8 experiment,After 24 hours,48 hours and 72 hours,the absorbance of breast cancer cells decreased gradually and was inversely proportional to the increase of concentration.Among them,10% of the compound group,20% of the compound group,and 10% of the positive group contained the drug serum acted on MCF-7 for 24 hours and 5% of the compound group contained the drug serum for 48 hours.Compared with the blank serum group at the same concentration,the difference was statistically significant(P<0.05);20% Salvia chinensia Benth group and 20% positive group contained the drug serum for 24 hours,the Salvia chinensia Benth group's three concentration groups(5%,10%,20%),10%compound group,20% compound group,and the positive group's three concentration groups(5%,10%,20%)acted for 48 hours and the three groups(5%,10%,20%)in the Salvia chinensia Benth group,the compound group,and the positive group had the effect of drug-containing serum for 72 hours,compared with the same concentration blank group at the same time,there was a significant difference(P <0.01)respectively.However,there was no significant difference between the 5% Salvia chinensia Benth group,5% compound group,5% positive group and 10% Salvia chinensia Benth group with the same concentration of the same concentration at the same time in the serum containing drug serum for 24 hours(P>0.05).2.The effect of Salvia chinensia Benth on mi R-21 and Bcl-2 expression Real-time PCR experiments: 48 hours after the administration of drug-containing serum of groups A,B,C and D in breast cancer cells MCF-7,the expression levels of miR-21 were different,and the expression levels from low to high were C,A,B,D.There was a significant difference in the expression level between A?B ?C group and D group(P<0.01).Western Blot experiments: 48 hours after the drug-containing serum of groups A,B,C,and D acted on breast cancer cells MCF-7,the expression of Bcl-2 was different.The expression levels from high to low were D,B,A,C.There was a significant difference in the expression level between A?B ?C group and D group(P<0.01).Conclusion:Salvia chinensia Benth can inhibit the proliferation of breast cancer cells and promote apoptosis of breast cancer cells,play an anti-tumor effect,and can down-regulate the expression of microRNA-21 and down-regulate the expression of Bcl-2 protein,suggesting that the anti-cancer mechanism of Salvia chinensia Benth is associated with downregulation of microRNA-21 and Bcl-2,there can inhibiting the proliferation of breast cancer cells and inducing apoptosis of breast cancer,playing an anti-cancer effect.
