Objective:Application of SBP(Salvia chinensis Benth.Polysaccharides) on humangastric cancer cell line (MGC-803cells) in vitro, to observe its influence oncancer metastasis, aiming to investigate whether the SBP(Salvia chinensisBenth.Polysaccharides) in vitro can inhibit the migration of gastric cancerMGC-803cells, as well as protein levels of interleukin-8(interleukin-8, IL-8)expressed in MGC-803cells when treated by SBP(Salvia chinensisBenth.Polysaccharides) in vitro. Further, provide a theoretical basis forclinical treatment of human gastric cancer.Methods:1Water extraction of SBP (Salvia chinensis Benth.Polysaccharides).Final concentration of the SBP (Salvia chinensis Benth.Polysaccharides)at100g ml-1.Culture of human gastric cancer MGC-803cells in vitro, withenvironments as follow: temperature at37℃,5%CO2incubator containing10%fetal bovine serum and green, streptomycin100U·ml-1, RPMI1640medium culture. Observation of cells`growth by inverted microscope,0.25%trypsin for digestion and passage, select the logarithmic growth phase cells forusage in the experiment.2(1) MTT colorimetric assay was applied to investigate the effects ofSBP(Salvia chinensis Benth.Polysaccharides)on the proliferative activity ofthe MGC-803cells. This experiment was repeated thrice.(2) Scratch-repair and Transwell experiments.Different concentrationsof SBP(0μg/ml,12.5μg/ml,25μg/ml,50μg/ml,100μg/ml)were used toculture gastric cancer in vitro. Scratch-repair and Transwell experimentswere used to detect cancer cell migrative ability, both of the experiment were repeated three times. Each experiment was done in triplicate and repeatedthrice.(3) Western blot was used to detect IL-8protein in gastric cancer cellMGC-803after treated by Different concentrations of SBP (0μg/ml,12.5μg/ml,25μg/ml,50μg/ml,100μg/ml) for24h respectively.This experimentwas done in triplicate and repeated thrice.3Statistical analysis: All data are expressed as mean±standard deviation(±S). SPSS17.0statistical software, statistical processing, the overall analysisof variance between groups were compared using LSD-t test. P <0.05wasstatistically significant difference.Results:1The impact of SBP(Salvia chinensis Benth.Polysaccharides) on thevalue-added activity of gastric cancer MGC-803cells when treated with25μg/ml,50μg/ml and100μg/ml concentration of scutellarin for24h,48h and72h respectively, the growth of MGC-803cells had showed time-dependentand dose-dependent manner. Through statistical analysis, the difference wasstatistically significant between groups (P <0.05).2The results of scratch-repair experiment shown that SBP(Salviachinensis Benth.Polysaccharides)could significantly decrease Cells`migrateinto the cell-free region in24h and the migrative ability was decreasedgradually with the dose increasing of SBP(Salvia chinensisBenth. Polysaccharides); The Transwell results shown that SBP(Salviachinensis Benth.Polysaccharides) significantly inhibit gastric cancer cells`invasion and cell number through Matrigel ECM membrane was deduced.There was statistical significance between different groups (P <0.01).3Western blot results showed that: protein expression levels of IL-8wasdecreased gradually with the dose increasing of SBP(0μg/ml,12.5μg/ml,25μg/ml,50μg/ml,100μg/ml)under the tested conditions. There wasstatistical significance between different groups (P <0.01).Conclusion:1SBP(Salvia chinensis Benth.Polysaccharides) inhibits proliferation, migration of human gastric cancer cell line MGC-803cells in vitro. Withincreasing drug concentration, the inhibition of cell proliferation, migration isenhanced.2Inhibition of the SBP(Salvia chinensis Benth.Polysaccharides) on theIL-8protein expression of human gastric cancer MGC-803cells. And with thedose increasing, the inhibition tend to strengthen.3The SBP(Salvia chinensis Benth. Polysaccharides) inhibites theexpression of IL-8in human gastric cancer MGC-803cells to achieve theinhibition of in vitro migration capacity.
