Objectives: Establishing the breast cancer complicated with depression animal model,and the model evaluation system by means of the behavioral,hematological indexes and hematological indexes,which laid the model foundation for the study of breast cancer complicated with depression.The mouse model of breast cancer complicated with depression was taken as the research object.The intervention of Xiaoyao Kangai Jieyu Compound was given to clarify the effect of anti-tumor and antidepressant.The mechanism of the treatment of breast cancer complicated with depression was preliminarily inferred,which provided a direction for the prevention and treatment of breast cancer complicated with depression.Methods: 1.Establishment of a model of breast cancer complicated with depression.BALB/c female mice were injected with 4T1 inflammatory breast cancer cells(107/ml,0.1 ml/L).7 days later,The mice were randomly divided into two groups according to the consumption of sugar water: breast cancer group and breast cancer complicated with depression group.Mice not injected with breast cancer cells were randomly divided into two groups according to the consumption of sugar water: blank group and depression group.Breast cancer complicated with depression and depression group were given subcutaneous injection of corticosterone 30 mg/Kg for 21 days to make depression model,and once a week to test the consumption of sugar water.After 21 days of making the model of breast cancer complicated with depression,the open field test and tail suspension test were measured in each group.The levels of plasma adrenocorticotr opin releasing hormone(ACTH),adrenocorticotropic hormone(ACTH),corticotropin 5-hydroxytr-yptamine(5-HT),serum dopamine(DA),norepinephrine(NE),carcino-marcinoma antigen(CA15-3),carcinoembryonic antigen(CEA)and carbohydrate antigen(CA125)were detected by Elisa.Detection of tumor weight and tumor volume.The pathological changes of hippocampus,tumor tissue and adrenal gland were observed by HE staining.The expression of NR2 A,NR2B,CREB and Ca MK? genes were detected by q RT-PCR..2.Intervention effect of Xiaoyao Kangai Jieyu Compound.The breast cancer complicated with depression mice model was established and randomly divided into six groups according to the consumption of sugar water: blank group,breast cancer complicated depression group,chemotherapy group,traditional Chinese medicine group,chemotherapy(20 mg/Kg)combined with antidepressant drug(2.6 mg/Kg),chemotherapy combined with traditional Chinese medicine group;delivered the at the time of depressive model.Results: 1.Compared with the normal group,In the breast cancer complicated depression group,the consumption of sugar water decreased significantly(P<0.01),the first feeding time in the novel feeding experiment increased significantly(P<0.05),the immobilit y time increased significantly in tail suspension test(P<0.01 or P<0.05).The expression of CA15-3,CA125 and CEA were significantly increased(P<0.01 or P<0.05);NE,DA and 5-HT were significantly decreased(P<0.01 or P<0.05;CORT,ACTH and CRH were significantly increased(P<0.01);the expression of NR2 A,NR2B,Ca MK? genes were significantly increased,the expression of CREB gene was significantly decreased(P<0.01).HE staining revealed obvious pathological changes in hippocampus and adrenal gland.2.Compared with the normal group,the indexes of the model mices of breast cancer complicated with depression were obviously abnormal.Compared with the model group,the consumption of sugar water and the independent behavior of Xiaoyao Kangai Jieyu Compound were significantly increased in mice,and behavioral despair improved significantly(P<0.01 or P<0.05);NE,DA and 5-HT were significantly increased(P<0.01);CORT,ACTH and CRH were significantly decreased(P<0.01);the expression of NR2 A,NR2B and Ca MK? genes were significantly increased,the expression of CREB gene was significantly decreased(P<0.01);Hippocampal injury and adrenal hyperfunction were alleviated.The expression of CA15-3 and CA125 were significantly decreased(P<0.01 or P<0.05),tumor weight and tumor volume decreased significantly in the BCRD group(P<0.01 or P<0.05).Conclusions: 1.The animal model of breast cancer complicated with depression was established by subcutaneous injection of corticosterone and axillary injection of breast cancer cells.2.Combined with the clinical characteristics of breast cancer complicated with depression,a series of indexes,such as sugar consumption,open field test,novel feeding test,tail suspension test,tumor markers CA153,CA125 and CEA,and the contents of monoamine neurotransmitter 5-HT,DA and NE were used to establish an animal model evaluation system for breast cancer complicated with depression.3.Xiaoyao Kangai jieyu Compound can obviously improve the general condition of mices with BCRD mice,rise the secretion of monoamine neurotransmitters.The therapeutic effects may be related to the protection of hippocampal injury,the reversal of HPA hyperfunction and the transformation of the expression of N R2 A,NR2B,Ca MK? and CREB genes.
