Gynostemma pentaphyllum(Thunb.)Makino is a perennial vines herbaeeous plant,belonging to the family of Cucurbitaceae,also known as Pentapanax leschenaultii.Gynostemma contains a variety of medicinal ingredients,such as saponins,flavonoids and polysaccharides,the whole plant can be use for medicine.Previous studies showed that Gynostemma pentaphyllum(Thunb.)Makino with the effects of hypolipidemic,blood pressure reduction and antioxidant activity.Gynostemma pentaphyllum belongs to dioecious,small amount of seed in natural conditions.In agricultral production,Gynostemma pentaphyllum depends on rhizome.Therefore,a large amount of seeds are waeted.It is very important to find the value of seeds.So,the study to identify seeds as the research object,its main ingredients grease is biology research,to improve the comprehensive development and utilization of Gynostemma pentaphyllum theoretical basis and technical support.In order to extract maximum Gynostemma pentaphyllum seed oil,the experiment of Gynostemma pentaphyllum was optimized by using orthogonal method and response surface method respectively seed oil ultrasound-assisted and microwave-assisted extraction technology and survey seed powder surface microstructure that after ultrasonic and microwave extraction residue powder and extracted without the original by using scanning electron microscopy.The GC-MS was used to analysis and compare Gynostemma pentaphyllum seed oil fatty acid composition that it from different solvent.Through the back of the neck subcutaneous injection of D-galactose mice aging model was established,the detection of serum and tissue of the physiological and biochemical indicators,to observe the mice liver biopsy,reserach the anti-aging function of Gynostemma pentaphyllum seed oil.The main results of the experiments are as follows:(1)Optimization of ultrasound-assisted extraction of seeds oil from Gynostemma pentaphyllum by orthogonal design,the optimal technological parameters were determined as follows:solid/liquid ratio of 1:20 g/mL,ultrasonic power of 300 W,ultrasound time of 40 min.Under these conditions,the yield of Gynostemma pentaphyllum seeds oil(GPSO)reached 40.941%,RSD 11%.The result showed that the extraction method was reasonable and reliable,achieved the goal of the optimization,which has some reference value.(2)Optimization of microwave-assisted extraction of seeds oil from Gynostemma pentaphyllum by response surface methodology,the optimal technological parameters were determined as follows:solid/liquid ratio of 1:15 g/mL,microwave temperature of 59 ?,microwave time of 14 min,microwave power of 600 W.Under these conditions,the yield of Gynostemma pentaphyllum seeds oil(GPSO)reached 42.0085%,which was close to the model predicted value 41.2459%.The result showed that the method was feasible and the optimized process was reliable.At the same time the seed residue powders that extracted by ultrasonic and microwave and the original seed powder observed by the scanning electron microscope.The results showed that the surface residue powder ware destroyed.In other words,ultrasonic and microwave extraction Gynostemma pentaphyllum seed oil were sufficient.(3)The GC-MS was used to analysis and compare Gynostemma pentaphyllum seed oil fatty acid composition that it from different solvent.The results showed that there were very large difference of Gynostemma pentaphyllum seed oil fatty acid from different extraction solvents.However it contains a lot of unsaturated fatty acid,especially the Conjugated linolenic acid.(4)The results of Gynostemma pentaphyllum seed oil anti-aging showed that each dose group can alleviate symptoms that diet decreased,surface glossed,weight slowed or declined.The serum biochemical TC,TG,LDL,HDL indicators that Gynostemma pentaphyllum seed oil can protect blood vessels so that it can compete aging.The tissue biochemical index show that Gynostemma pentaphyllum seed oil can increase SOD activity significantly and reduce MDA activity,increase total antioxidant capacity T-AOC.It can say that Gynostemma pentaphyllum seed oil is one of the anti-aging mechanism may be related to its antioxidant ability,strengthen the body's total antioxidant capacity and effectively remove free radicals,reduce the body's cells damage degree,indirect protection of normal cells,which have the function of anti-aging.
