| Purpose:To compare and optimize the extraction and purification techniques of total saponins in Gynostemma pentaphyllum(Thunb)Makino;To study the chemical constituents of the hydrolysate of the saponins from Gynostemma pentaphfllum;To determinate the quantitative of new compounds from the hydrolysates of the Gymostemma pentaphyllum.Material and method:Took the content of total saponins in Gynostemma pentaphyllum as mark,determine and compare the content of total saponins that was determined by spectrophotometric method through the different extraction,and compare the static and dynamic adsorption and elution capability of total saponin with six different macroporoils resins;The compounds isolated were separated by repeated silica gel chromatographies.Their structures were identified by analysis of their spectral data;Using high performance liquid chromatography (HPLC) to determinate the quantitative of new compounds from the hydrolysates of the Gymostemma Pentaphyllum.Results:1.The content of total saponins by Smashing Tissue Extraction was 32.69%,the content of total saponins by ethanol refluxing was 31.19%;HPD-700 had the highest eluted ratio of 80.34%.2.9 compounds were isolated from the transformed products and 5 compounds have identified on the basis of physico-chemical and spectrographic evidence,there were 20(S)-panaxadiol(â… ),20(R)-protopanaxadiol(â…¡),20(R)-dammarane-3β,12β,20,25-tetrol(â…¢), JGL-81(â…£),JGL-YY-1(â…¤).3.In the hydrolysates of the GymostemmaPentaphyllum,the quantitative of JGL-81 was 5.86%,JGL-YY-1 was 1.16%.Conclusion.1.The method of using Smashing Tissue Extraction to extract and purified by HPD-700 macroporous absorption resin was a feasible extraction and purification process of total saponins in Gynostemma pentaphyllum. 2.Compoundsâ…£andâ…¤were new compounds.3.Using high performance liquid chromatography determinate the quantitative of JGL-81 and JGL-YY-1,the results showed that the linear ranges of the standard curves were 2.0-10.0μg. This method has the good isolation effect for the hydrolysates.Meanwhile,it has the feature of fast,simple and good reproducibility. |
