The Traditional Chinese Medicine(TCM)of seed of the Peganum harmala L.plant.Bitter,warm,toxic,with antitussive and antiasthmatic,rheumatism,pass through and other effects.Harmine has a series of pharmacological effects such as antibacterial,anti-parasite,anti-fungal,anti-oxidation,anti-tumor,anti-mutagenic,cytotoxic and hallucinogenic.In order to better study the composition changes before and after the processing of camel pawbush,clarifying the processing mechanism of camel beetle,this article mainly carried out the content determination and activity evaluation of different processed products of camel bengalensis,simulated processing research and harmonine and derivatives thereof.The study of synthesis and activity evaluation provides a basis for the study of processing technology and quality control.1.Identification of alkaloids in seeds from seed of peganum harmala by HPLC-MSThe extract of seeds of P.harmala were analyzed using high performance liquid chromatography coupled with high-resolution mass spectrometry.Chromatographic separation was achieved on a C18 column with the mobile phase of acetonitril-water(0.5%ammonia water)and step gradient elution.The ESI-MS analysis was operated at the mass range of m/z 100-1000 under the positive ionization modes,respectively.A total of 30 compounds of seeds from were identified from the methanolic extract in the seeds of Peganum harmala.,such as 10isoquinoline alkaloid and 11 quinazolone alkaloids,which are characterized in the methanolic extract,based on the accurate mass of molecular and product ions provided by LTQ-Orbit rap mass spectrometers.As far as we know,these compounds have been recognized in seeds of Peganum harmala for the first time:glycosides vasicine,glycosides pegamine,3-hydroxylated harmine The method is rapid and simple in the qualitative analysis of licorice using HPLC-LTQ-Orbit rap MS,which could proves to have a high sensitivity and the result is accurate and reliable.It is an effective method to provide chemical information concerning the constituents in herbal medicines,which could be helpful for the quality control and further phytochemical studies of seeds of Peganum harmala L.2.Study on content determination and activity evaluation of different processed products of seed of peganum harmalaHPLC method was used to determine the content of harmaline and harmine in the processed products of seed of peganum harmala.The Muv phenotype of let-60(n1046f)mutant and the human pancreatic cancer cell line PANC-1 were used as models for The activity of different processed products was evaluated.The results showed that the HPLC method can effectively separate the harmaline and the harmine.And the processing of seed of peganum harmala,the content of the harmaline from high to low is as follows:seed of peganum harmala?wine seed of peganum harmala>vinegar seed of peganum harmala>fried seed of peganum harmala,the content of harmine from high to low is:fried seed of peganum harmala>vinegar seed of peganum harmala>wine seed of peganum harmala>seed of peganum harmala.Seed of peganum harmala of different processed products for two models of different processed products has inhibitory effect.3.The simulation processing of seeds of Peganum harmala L.The HPLC method was used to compare the content of the harmaline in water heated at 95? for different time,and heated in 95? acid water for different time.The decomposition of the harmaline can be carried out by hydrolysis,acid hydrolysis,and baking,and the harmine can be desorbed by removing one hydrogen atom from each of the 3 and 4 positions of harmaline.The hydrolyzed content of harmaline gradually decreased.The hydrolysis rate increased significantly after 10 minutes of acid hydrolysis,while the hydrolysis rate increased after 20 minutes,indicating that the hydrolysis rate of marigold was increased after the addition of acid;In the generation of harmine,with the heating temperature,time,the main factor is the temperature.With the increase of temperature,the harmaline content is more significant.4.Quantum chemical calculations of the decomposition reaction of harmalineAccording to the results of simulation processing of seeds of Peganum harmala L.,the structure of harmaline and harmine was optimized using the DMol3 module of Materials Studio 2017 and the B3LYP method of Gaussion07,and the decomposition process of harmaline was calculated using Materials Studio 2017 software.The results show that the energy required to decompose to a transition state of harmaline is 3.569 kcal/mol,which is less than 4.369 kcal/mol from the transition state to the harmine and hydrogen,indicating that the reaction is more likely to occur in the forward direction and the reverse reaction is more difficult5.The synthesis and activity evaluation of harmolHarmol was synthesized from harmine as the raw material.The activity of harmol was evaluated based on the Muv phenotype of let-60(n1046gf)Caenorhabditis elegans model and human retinal vascular endothelial cells.The results showed that harmol had inhibitory effects on the MvV phenotype of the let-60(n1046gf)Caenorhabditis elegans and human retinal vascular endothelial cells,but its inhibitory effect on the two models was significantly weaker than that of harmine.In addition,the minimal non-toxic dose of harmol to HRCEC cell lines was significantly higher than that of harmine,confirming that the 7-position oxoalkyl group of harmine was reduced in cytotoxicity as the carbon chain decreased,which provided the basis for the study of the salmonine derivatives.
