Objective:To observe the effect of moxibustion on CD3-CD161+NK and CD3+1CD161+ NKT in peripheral blood of tumor bearing gastric carcinoma rats.To explore the therapeutic effect and possible mechanism of moxibustion on gastric cancer,and to provide experimental basis for clinical application of moxibustion in treating gastric cancer.Methods:50 healthy SD rats were randomly divided into 5 groups:blank group,sham-operated group,model group,moxibustion group and infrared group,10 rats in each group,male and female half.Walker-256 cancer cells were used to prepare gastric carcinoma model in model group,moxibustion group and infrared group by situ implantation of tumor tissue.After model succeed,suspended moxibustion was given to moxibustion group.Acupoints were divided into two groups:zhongwan,guanyuan and zusanli for one part,pishu and weishu for the other.And two groups of acupoints were treated with moxibustion alternately.Short wave infrared radiation was applied to infrared group on the epigastrium and lower back,which were the surface projection areas of stomach,while the rest groups were bundled merely.Operation were carried out one time per day and 20 minutes per time for 21 days in all groups.The general symptom scores were recorded in the model group,moxibustion group and infrared group during whole treatment.At the end of the treatment,orbital blood of rats in each group was collected,and flow cytometry was used to measure content of CD3-CD161+NK and CD3-CD161+NKT in peripheral blood.The stomach,thymus,spleen and stomach tumor tissues of rats in each group were removed.We measured the tumor volume to calculate the tumor growth inhibition rate.The thymus index and spleen index were determined.Results:1.After tumor formation,the general symptom scores decreased with time in model group,moxibustion group and infrared group.10 days,15 days and 20 days after tumor formation,the score of model groups were lower than blank group and sham-operated group(P<0.05).The score of moxibustion group was higher than that of the model group(P<0.05).15 days and 20 days after tumor formation,the score of moxibustion group was higher than infrared group(P<0.05).2.The volume of gastric tumor in moxibustion group and infrared group was smaller than the model group(P<0.05).The volume of gastric tumor in moxibustion group was smaller while the growth inhibition rate of gastric tumor was apparently higher than in infrared group(P<0.05).3.Compared to sham-operated group,the spleen index of model groups were increased(P<0.05).Compared to model group,the increase of spleen index and thymus index was apparently in moxibustion group(P<0.05).And the moxibustion group was superior to infrared group(P<0.05).4.Compared to blank group,the CD3+CD161+NKT content in the peripheral blood in model groups all increased(P<0.05).Compared to model gro-up,the CD3-CD161+NK and CD3+CD161+NKT content in the peripheral blood of moxibustion group was increased(P<0.05).Compared to infrared group,the CD3+CD161+NKT content in the peripheral blood in moxibustion group increased more apparently(P<0.05).Conclusions:1.It can effectively improve general symptoms and survival state of the gastric tumor rats by treating with moxibustion.2.Moxibustion and infrared treatment both can reduce the volume of gastric tumor,and moxibustion has better inhibitory effect on gastric tumor growth.3.Moxibustion can significantly increase the content of CD3-CD161+NK and CD3+CD161+NKT in peripheral blood,improve thymus index and spleen index of tumor-bearing rats.And moxibustion improved the survival state of rats.The regulation mechanism is related to the strengthening of non-specific anti-tumor immunity.
