Objective:To analyze the frequency of CD161~~+CD4~~+T cells,INF-γ~+CD4~+CD161~+T cells, IL-22~+CD4~+CD161~+T cells in peripheralblood mononuclear cells (PBMCs) from rheumatoid arthritis (RA) patientscompared with healthy control subjects,(1) To discuss the relationship ofCD4~+CD161~+T lymphocyte frequency changes with RA disease activity, andto further explore the pathogenesis of RA, and to look for new treatments ofRA in the future;(2) To study the role that CD4~+CD161~+T lymphocytes playin RA bone destruction.Method:21cases in RA group, all of them were the inpatients oroutpatients in the Tangshan Gongren Hospital Rheumatology Departmentfrom2010to2011. All21cases included14female and7male, age36-74years old, average (55±10ys), the average course (60±50ms), are in line withthe American College of Rheumatology (American College of Rheumatology,ACR) classification diagnosis standard.21cases in control group, all of themhad health examination in Tangshan Gongren Hospital,15female and6male,age40-78years old, average age (55±9ys). CD4~~+CD161~~+T cells in PBMCswere detected by flow cytometry. The frequency variation of differentCD4~~+CD161~~+T were compared between case and control groups.Thecorrelation was studied between the frequency and RA disease activity score(DAS28), tender joints, swollen joints, red blood cell sedimentation rate,high sensitivity C reactive protein and white blood cell, red blood cell, platelet,IgG, IgA, IgM, complement C3, complement C4and so on.Result:(1) CD4~~+T lymphocyte number (45.85±10.61) in peripheralblood mononuclear cells in RA group were significantly higher than those ofhealthy controls (35.49±8.10), the difference was statistically significant (P=0.001); CD4~+CD161~+T lymphocyte number in control group (27.53±7.34) was higher than that in RA group(23.32±5.97), the difference was statisticallysignificant (P=0.048); CD4~+CD161~+T cell and CD4~+CD161T cell (0.39±0.15), CD161~+CD4T cell (46.66±11.91) in controls were higher than that inRA group [(0.32±0.14),(39.42±11.83)], but without statistical significance[P=0.078, P=0.158].(2) The secretion INF-γCD4~+T cell in RA group(11.06±6.68) was higher than those in the control group (9.42±3.54), but nostatistical significance (P=0.678); The secretion INF-γCD4~~+CD161~~+Tcells(11.3±4.35), the secretion INF-γCD4~+CD161~+T cells/the secretionINF-γCD4~~+CD161~~+T cells (0.76±0.29) in controls were higher than that inRA group (8.22±3.94),(0.59±0.3)], and the difference was statisticalsignificance [P=0.021, P=0.048];(3) The secretion INF-γ and IL-22CD4~+T cells (0.33±0.20), the secretion IL-22CD4~~+T cell (0.51±0.29), thesecretion IL-22CD4~+CD161~+T cell (0.55±0.28) in RA group at the same timewere significantly higher than that in control group [(0.22±0.14),(0.25±0.18),(0.36±0.24)], and were statistically significant different [P=0.045, P=0, P=0.016].Conclusion: CD4~+CD161~+T lymphocyte count in peripheral bloodmononuclear cells in RA group were significantly lower than those in controlgroup, suggesting that CD4~~+CD161~~+T lymphocytes in RA patients may havemigrated into the joint. IL-22~~+CD4~~+Tcells in RA patient peripheral bloodmononuclear cells was significantly higher than that in control group, IL-22isconnected with the activity of RA.
