Objective:To study the effect of Bushen Huayu Recipe on serum vitamin D3,AMH,T,INS and E2 in rats with polycystic ovary syndrome?PCOS?and the changes of endogenous metabolites in ovarian tissues after intervention with Western and Chinese medicine.We analyzed the changes of serum indexes of rats in each group after drug intervention,explored the mechanism of the improvement of PCOS.According to the difference of ovarian tissue metabolites,We found out the relative metabolic pathways involved and potential biomarkers,In order to further explore the mechanism of this prescription in the treatment of PCOS.Methods:The eight of 61 SD female rats were randomly selected as blank groups.The remaining rats by gastric perfusion of letrozole to establishment of PCOS rat model,which rats were randomly divided into the model group,the Chinese medicine group and the western medicine group.The blood of abdominal aorta was taken after the intervention of the corresponding drug intervention.The serum levels of AMH,INS,T,25?OH?VD3 and E2 in each group after with drug intervention were measured by enzyme-linked immunosorbent assay?ELISA?through analysis of the difference of serum levels in each group.At the same time,the UPLC-MS technique was used to analyze the metabolomics of the ovarian tissue of PCOS rats after drug intervention.The selected metabolites could serve as potential metabolic markers of PCOS and construct relative metabolic pathways.Results:The results of serum index is that the concentrations of serum E2 and 25?OH?VD3 in the blank group,the Chinese medicine group and the western medicine group were higher than those in the model group,the results were significant difference?P<0.05?.While the serum AMH and INS concentrations among three groups were lower than those in the model group,which were significant difference too?P<0.05?.Metabolomics study results showed that the model group of ovarian tissue metabolites such as N1,N10-Diferuloyl spermidine,Nicotine and PG?18:3?9Z,12 Z,15Z?/20:4?5Z,8Z,11 Z,14Z??levels decreased compared to the blank grou.While other metabolites,such as Ajoene,Thioxanthine monophosphate,Diethyl sulfate,Methyl 2,6-dihydroxy-4-quinoline carboxylate,L-Anserine,etc.The blank group revealed in a rising state.Through the differential metabolites screened after drug intervention,the most relevant metabolic pathways for the treatment of PCOS were constituted by Bushen Huayu Recipe,which were cytochrome P450 exogenous material metabolism pathway,?-alanine metabolic pathway and histidine metabolic pathway.Conclusion:Bushen Huayu Recipe can regulate the levels of AMH,INS,25?OH?VD3 and E2in the serum of polycystic ovary syndrome?PCOS?rats by letrozole,which can effectively improve PCOS's Anovulatory,follicular development stagnation and disorder of glycolipid metabolism.Bushen Huayu Recipe can also regulate cytochrome P450 exogenous substance and amino acid metabolism pathway in PCOS rats to improve the imbalance of body metabolites.
