| Objective:This research is to observe the effect of Xintongtai on the expression of Toll like receptor 4(TLR4),p38 mitogen activated protein kinase(p38 MAPK),myeloperoxidase(MPO),Ras gene family member A(RhoA),endothelial nitric oxide synthase(eNOS)in the rabbit model of atherosclerosis and elucidate the possible treatment mechanism of Xintongtai on atherosclerosis.Methods:120 rabbits were randomly divided into control group,model group,high dose group,median does group,low dose group,rosuvastatin group with 20 rats in each group.Except the control group,the other groups were fed with high-fat diet combined with immune injury to establish a rabbit model of atherosclerosis,while daily treatment of Xintongtai.Low,medium and high dose group was respectively given 2.3g/kg,4.6g/kg,9.2g/kg.The model group and normal control group were given equally volume of physiological saline.The treatment is a total of s weeks.After the end of the experiment,the pathological condition of rabbit aortic tissue was detected by HE staining.The expression levels of TLR4,p38 MAPK,MPO,RhoA,eNOS protein and mRNA were detected by Western blot and RT-PCR.SPSS 19.0 software was used for statistical analysis of data.Results:Compared with control group,the level of TLR4,p38MAPK,MPO,RhoA mRNA and protein were increased significantly,while the level of eNOS mRNA and protein were decreased(P<0.01).Compared with model group,the level of TLR4 mRNA and protein,p38MAPK protein in low does group were decreased(P>0.05),the level of MPO,RhoA mRNA and protein were decreased(P<0.05),while the level of p38MAPK mRNA was decreased and the level of eNOS mRNA and protein were increased in low dose group(P<0.01).Compared with model group,the level of TLR4 mRNA was decreased(P<0.05),and the level of p38MAPK,MPO,RhoA mRNA and protein were decreased,the level of eNOS mRNA and protein were increased in median does group(P<0.01).Compared with model group,the level of TLR4、p38 MAPK、MPO、RhoAmRNA and protein were decreased,while the level of eNOS mRNA and protein were increased in high dose group and rosuvastatin group.There is no significant change between the rosuvastatin group and the high-dose group(P>0.05).Conclrusion:Xintongtai can decrease the expression of TLR4,p38 MAPK,MPO,RhoA protein and mRNA,up regulate the expression of eNOS protein and mRNA,inhibit inflammation in blood vessels,and protect the vascular endothelium of aorta,so as to achieve the aim of treating atherosclerosis. |
