Objective:Discussion the mechanism of the Xintongtai influence TNF-α,IL-1level and the expression of NF-kappa B in atherosclerotic mice.Methods: A total of 42 6-8 week old male Apo E-/-mice fed a high-fat diet feeding,another 12 mice as control group,fed with normal diet.After 12 weeks of feeding,from two mice randomly selected 2 rats,HE staining observation shows that the Apo E-/-mice aortic high fat diet has obvious instability of atherosclerotic plaques and the formation of foam cells,and the control group of wild-type mice fed with normal diet in aortic smooth membrane preparation showed that the AS model success(Figure 1).40 Apo E-/-mice were divided into model groups according to the random number table method,and the low,high dose group and atorvastatin calcium group were added to the group,and the other group was added to the blank group.Were given saline,low dose and high dose Xintongtai and Atorvastatin Calcium Tablets 8weeks later,selected samples of animal,under sterile conditions,the total serum cholesterol was determined by enzyme method(TC),triglyceride(TG),the determination of low density lipoprotein cholesterol assay(LDL-C),high density lipoprotein protein cholesterol(HDL-C),aortic endothelial cell morphology and pathological changes were observed under light microscope,ELISA assay of serum TNF-,IL-1 levels were detected in nucleus and cytoplasm of NF-in mice aorta B kappa p65 protein expression by Western blot method,to detect the expression of mouse aortic endothelial cells NF-kappa B m RNA RT-PCR method.Results: 1.compared with the model group,the drug groups can reduce the serum levels of TC,AS TG,LDL-C,HDL-C increased,and the Xintongtai dose group than in low dose group(P<0.05),but the Xintongtai dose group and atorvastatin group have no difference(P>0.05).2.Compared with the model group,the drug groups could decrease AS serum level of TNF-α and IL-1 level(P<0.05),and Xintongtai high dose group,atorvastatin calcium group is better than the low dose group(P<0.05).3.Compared with the model group,the expression of the drug group AS miceaorta nuclear NF-κB p65 decreased significantly(P<0.05);and the Xintongtai high dose group,atorvastatin calcium group aortic nuclear NF-κB p65 were significantly lower than those of Xintongtai low dose group(P< 0.05).4.Compared with the model group,the drug group AS mice aorta NF-κB p65 protein expression increased significantly(P <0.05),and Xintongtai high dose group,atorvastatin calcium group aortic cytoplasmic NF-κ B p65 protein is significantly better than the Xintongtai low dose group(P < 0.05).5.Compared with the model group,the expression of the drug group AS mice aorta NF-κB m RNA decreased significantly(P<0.05);and the Xintongtai high dose group atorvastatin calcium group aortic NF-κ B m RNA was lower than the Xintongtai low dose group(P < 0.05).Conclusion:1.Xintongtai granules could down regulate the serum TC,TG and LDL-C of AS mice and up regulation of HDL-C.It indicated that Xintongtai granule could interfere with the development of AS by regulating blood lipid.2.The observation of light microscope showed that Xintongtai granules could improve the histopathological changes of the endothelial cells of AS mice and reduce the lipid infiltration and the formation of foam cells and plaque.3.Xintongtai granules can reduce the level of TNF-α and IL-1 in the serum of AS mice,thus reducing the release of inflammatory mediators and interfering with the AS process.4.Xintongtai granules can inhibit the transposition of NF-κ B p65 nucleus and the expression of NF-κ B m RNA in the aorta of AS mice,inhibit the inflammatory response and interfere with the progress of AS.
