Study On Some Chemical Constituents And Active Diversity Of Dichloromethane Of Polygonum Vulgare

La Liao（Poygonum hydropiper L.）,which is also called Water Liao,belongs to Polygonaceae Polygonum."Persicaria hydropiper"is another famous synonym for the species.La Liao is distributed in worldwide with a wide range of traditional uses.It plays a great role in the long history.La Liao is used in the treatment of gastroenteritis,dyspepsia,diarrhea,itching skin and so on.Until now,many literatures have reported Polygonum hydropiper is rich in chemical ingredients,flavonoids,sesquiterpenoids,steroids,phenylpropanoid esters,tannins and volatile oils,etc.And it has a wide range of activities,anti-oxidation,antibacterial,deworming,anti-inflammatory agents,anti-cancer,anti-rheumatism and so on.However,as a curative effect and widely used traditional Chinese medicine,La Liao’s chemistry and activity research are not thorough,the sources are confused,and the quality standards are imperfect.The phytochemical studies of dichloromethane extracts from Polygonum hydropiper have led to the isolation of 14 compounds by silica gel column chromatography（CC）,sephadex LH-20 CC,recrystallization and the identification of them by spectroscopic analysis and comparison with literatures.There were vanicoside B（1）,vanicoside F（2）,vanicoside E（3）,isorhamnetin（4）,3,7-dihydroxy-5,6-dimethoxy-flavone（5）,3,5-dihydroxy-7-methoxy-flavone（6）,2’,4’-dihydroxy-6’-methoxychalcone（7）,pinosylvin（8）,5,6-dehydrokawain（9）,aniba-dimer-A（10）,6,6’-（（1R,2R,3S,4S）-2,4-diphenylcyclobutane-1,3-diyl）bis（4-methoxy-2H-pyran-2-one）（11）,（+）-ketopinoresinol（12）,2-desoxy-4-epi-pulchellin（13）,β-sitosterol（14）.Compounds 5,6,12,13 were firstly obtained from Polygonum hydropiper.Compound 7,9-11 were firstly isolated from Polygonum.And compounds 2 and 8were firstly report in polygonaceae.In this study,the active screening test was carried out for Polygonum hydropiper extract layers and isolated compounds（sufficient amount of activity test compounds）.The biological activities are antitrypanosomal,antimalarial,antimicrobial,anti-inflammatory,PPAR and cytotoxic assays.Polygonum hydropiper extracts and many compounds were found to show excellent activity results.Especially compound2’,4’-dihydroxy-6’-methoxychalcone（7）is the most active one with IC₅₀/IC₉₀values of0.49/0.81μg/m L,which is more potent than the positive control alpha-difluoromethylornithine（DFMO）with IC₅₀/IC₉₀values of 3.02/8.05μg/m L.The above works were effort to the novel,safe and effective drug discovery from therapeutic plants.
