Purpose: To evaluate the effects of collagen type I-PRP composite on promotingfunctional differentiation of tendon stem cells(TDSCs).Methods:①TDSCs were isolated form rabbit patellar tendon and Achillestendon. The isolated TDSCs were characterized by immunostaining for stem cellmarkers and characterized for its multi-differentiation potential.②TDSCs at passage2were cultured on collagen I-PRP scaffold. After3days culture, quantitativereal-time PCR (qRT-PCR) was used to characterize the mRNA expression ofCollagen type I, Tenascin C, Oct-4, Nanog and immunohistochemical analyses wereused to characterize the protein expression of Nucleostemin, Oct-4, Tenascin C.③TDSCs were mixed with collagen I-PRP composite and implanted subcutaneouslyinto the abdominal areas of nude rats. After2weeks, HE staining was used to test theorganization of the tissue and immunohistochemical analyses were used to test theprotein expression of collagen types I and CD31.Results:①The isolated TDSCs had a multi-differentiation potential andpositive staining of the stem cell markers.②TDSCs in collagen I-PRP scaffold grouphad more Tenascin C expression (p<0.05). There was no statistical differencebetween collagen I-PRP scaffold group and control on Collagen type I, Oct-4, Nanogexpression (P>0.05). Immunohistochemical analyses showed that TDSCs in collagenI-PRP scaffold group secreted more Tenascin C compared to control group.③TDSCs mixed with collagen I-PRP composite formed tendon-like tissue after2weeksculture. TDSCs in collagen I-PRP composite group secreted more collagen type I andCD31.Conclusions:①The isolated TDSCs have a multi-differentiation potential andpositive staining of the stem cell markers.②TDSCs can maintain their stemnesswhen cultured on collagen I-PRP scaffold and can express more tendon related genes. ③Collagen I-PRP scaffold can promote TDSCs to form a tendon-like tissue, tosecrete more tendon related protein and promote formation of new vessels.
