Research purposesA rat model of damp-heat hyperuricemia was established.Through the intervention of traditional Chinese medicine,the effect of Tongfeng No.2 on blood uric acid concentration,serum creatinine,blood urea nitrogen,and serum and renal nitric oxide and endothelin-1 levels were observed.A comparative analysis of western medicine was conducted to assess the effect of Gout II on the control of hyperuricemia and the intervention of renal damage.Research methods:Experimental research methods: randomly divided into blank group,model group,allopurinol group,gout 2nd group.The blank group: normal feeding,the same amount of saline gavage.Model group: Preparation of a hyperuric heat and humidity model.Gout No.2 group: Rats were given gavage on the second day of gout intervention until the 14 th day.Allopurinol group: Allopurinol(1 mg/100 g)was intragastrically administered to rats until the 14 th day.The serum uric acid concentration in rats was measured by biochemical instrumentation.Serum uric acid,serum creatinine,blood urea nitrogen,serum and renal nitric oxide,and hematopoietin-1 levels were measured by enzyme-linked immunosorbent assay;kidneys were observed by HE staining.The pathological changes.Research result:(1)Serum uric acid index(SUA): The serum uric acid concentration in the model group was significantly higher than that in the control group(P<0.01).In addition,the allopurinol group and the model group and gout There was a significant difference between the two groups and the model group(P<0.01),and the difference between the two groups was extremely significant(P<0.01).(2)Observed by light microscope: Compared with the blank group,the model group has obvious changes,mainly seen with glomerular enlargement,balloon adhesion,narrowing of the renal cyst,or disappearance,and protein tube type seen in the renal tubules.It indicates that the kidney has severe histopathological damage;the Chinese medicine group and allopurinol group,glomerular enlargement,balloon adhesion,renal tubular epithelial cells to varying degrees of swelling,the above pathological damage degree is slightly smaller than the model group,more close to the blank Group,and there was no significant difference between the two treatment groups.(3)Serum creatinine index: The serum creatinine level in the model group was higher than that in the blank group(P<0.01).The serum creatinine level was significantly higher in the model group and the Tongfeng No.2 group.There was significant difference(P<0.05)between the allopurinol group and the model group(P<0.05).There was a significant difference between the drug groups(P>0.05).(4)Urea nitrogen(BUN)index: Compared with the blank group,the BUN concentration in the model group was significantly higher(P<0.01),and the model group and the Gout II group and the model group were different.There was a significant difference between the sterol groups(P<0.05),and there was a significant difference between the drug groups(P<0.05).(5)Endothelin-1(ET-1)index: 1 Serum ET-1: Compared with the blank group,the blood ET-1 concentration in the model group was significantly increased(P<0.01),gout There was a significant difference between the No.2 group and the model group(P<0.01),and there was significant difference between the allopurinol group and the model group(P<0.05),and there was no significant difference between the allopurinol group and the Gout No.2 side(P> 0.05);2 Renal tissue ET-1: model group and blank group,allopurinol group,Gout 2nd group have extremely significant differences(P <0.01),while there is no significant difference between the drug group(P >0.05).(6)Nitric oxide(NO)index: 1 NO in serum: Compared with the blank group,NO concentration in the blood of the model group was significantly reduced(P<0.01),Gout No.2 group and allopurinol There was a significant difference between the group and the model group(P<0.01).There was a significant difference between the allopurinol group and the Gout No.2 side(P<0.05);2 The NO of the kidney tissue: the model group and the blank group,allopurinol group There was a significant difference between the Chinese medicine group and the Chinese medicine group(P<0.01),but there was a significant difference between the Chinese medicine group and the allopurinol group(P<0.05).Conclusion: 1.Gouty No.2 and allopurinol have preventive and therapeutic effects on damp-heat hyperuricemia in rats,and Gout No.2 is superior to allopurinol 2.Gout 2 has less acute toxicity to renal function and is a safer formula.Its effect is better than that of allopurinol.3.The mechanism of improvement of renal function damage by Gout No.2 may be related to the increase of renal NO level and the decrease of ET-1,BUN,and Cr...
