Objective: Observing the acupuncture combined hypothermia on cerebral ischemia reperfusion injury(CIRI)in rats model of nerve function defect grade and area of cerebral infarction,MAPK pathway Cacnb2,Epha4 and Arrb2 level as well as the influence on mi RNA181,explore the effect of acupuncture combined hypothermia therapy for ischemic stroke mechanism.Methods: by reference to Zea Longa line switch method and modified method,the replication MCA ischemia model(MCAO),60 SD rats were randomly divided into blank group and control group,model group,acupuncture group,the low temperature and acupuncture combined hypothermia group 6,72 h after treatment in a row,neural function defect scale in MCAO rats;TTC staining to detect infarct area;PCR fluorescence quantitative analysis used to detect the levels of mi RNA181,Cacnb2,Epha4 and Arrb2 in rats with ischemic lateral hippocampal tissue.Results:1,.Neural function defect scale comparison: before the treatment,each building module(model group,acupuncture group,the low temperature group,the acupuncture combined hypothermia group)compared with the blank group and control group,building module neural function defect scale increases,statistically significant difference(P <0.01);The difference between each model group was statistically significant(P>0.05),indicating the success of the model.72 hours after treatment,compared with model group,the treatment group(acupuncture group,the low temperature group,the acupuncture combined hypothermia group)scores than the lower,with significant difference(P<0.01).2.Cerebral infarction area ratio comparison: compared with the blank group and control group,building module infarction area of rats were significantly increased remarkably,with significant difference statistically significant(P <0.01),which indicates the success of model;After 72 h treatment,compared with the model group,the infarct area of each treatment group was significantly reduced,and the difference was statistically significant(P<0.01.).Compared with the acupuncture group and sublow temperature group,the ratio of the infarcted area of the acupuncture union subgroup was significantly reduced,and the difference was statistically significant(P<0.01).3.Comparison of mirna-181 expression in brain tissue: compared with the model group,the expression of mnirna-181 in the brain tissue could be reduced by each treatment group,and the difference was statistically significant(P <0.01).Compared with the acupuncture group and sublow temperature group,the expression of mirna-181 was significantly down-regulated by acupuncture combined with subcryogenic group,but the difference was not statistically significant(P>0.05),which may be related to the insufficient sample quantity.4.Expression of Cacnb2,Epha4 and Arrb2 in ischemic hippocampal tissue in each group:Compared with the model group,the expression of Cacnb2 in brain tissue could be down-regulated by acupuncture group,subhypothermia group,acupuncture combined with subgroup,and the difference was statistically significant(P<0.01).Compared with sublow temperature group,the expression of Cacnb2 was significantly reduced by acupuncture combined with subcryogenic group,and the difference was statistically significant(P<0.01).Compared with the model group,the expression of Epha4 in the brain tissue could be reduced by acupuncture group,subhypothermia group,acupuncture and subhypothermia group,and the difference was statistically significant(P<0.01).Compared with hypothermia group and acupuncture group;The expression trend of Epha4 was significantly reduced by acupuncture combined with subcryo group,but the difference was not statistically significant(P>0.05),which may be related to the insufficient sample quantity.Compared with the model group,the expression of Arrb2 in brain tissue could be reduced by acupuncture group,subhypothermia group,acupuncture combined subgroup,and the difference was statistically significant(P<0.01).Compared with acupuncture group and sublow temperature group,the reduction of Arrb2 expression in acupuncture combined with subcryogenic group was obvious,but the difference was not statistically significant(P>0.05),which may be related to the insufficient sample quantity.Conclusion:1.the acupuncture group,mild hypothermia group,acupuncture group and acupuncture combined with hypothermia group can reduce the neural function defect scale of MCAO rats,to a certain extent reduce the cerebral infarction area,at the same time can play a protective effect on brain tissue.2.The protective mechanism of acupuncture combined with hypothermia for brain tissue may be implemented by down-regulating mi RNA181,Cacnb2,Epha4 and Arrb2 levels on P38 MAPK pathway.
