Objective:By observing the acupuncture combined mild hypothermia therapy of Middle cerebral artery ischemia in rats of neural function defect scale,cerebral infarction area ratio,in brain tissue of mir-29 c and its target genes Map4k4,Map3k14,Fem1 b expression,acupuncture combine the low temperature of brain tissue damage caused by cerebral ischemia reperfusion protection mechanism.Methods: Using modified Zea Longa line tied the preparation of MCAO rats model,60 SD rats were randomly divided into blank group and control group,model group,acupuncture group,the mild hypothermia group,the acupuncture combined mild hypothermia group,10 in each group.Acupuncture group,the mild hypothermia group,acupuncture combined hypothermia group acupuncture,hypothermia,acupuncture combined with mild hypothermia treatment,after 72 hours,observe each rat neural function defect scale,bilateral hippocampal tissue ischemia,cerebral infarction area ratio of mir-29 c and its target genes Map4k4,Map3k14,Fem1 b expression.Results:1.Nerve function score: before the treatment,each building module(model group,acupuncture group,the low temperature group,the acupuncture combined hypothermia group)compared with the blank group,the control group,the neurological function score building group were significantly increased,with significant difference statistically significant(P<0.01),compared between each building modules,there was no statistically significant difference(P>0.05),shows that building success.After treatment of 72 h,the scores of each treatment group(acupuncture group,mild hypothermia group,acupuncture combined mild hypothermia group)were significantly decreased compared with the model group,and the difference was statistically significant(P<0.01).2.Ratio of cerebral infarction area: compared with the blank group and the sham group,the ratio of the infarct area of the model group increased significantly,and the difference was statistically significant(P<0.01),indicating that the model was successful;After 72 h treatment,compared with the model group,the ratio of infarcted area in each treatment group decreased,and the difference was statistically significant(P<0.01).Compared with acupuncture group and mild hypothermia group,the ratio of infarcted area in acupuncture combined with mild hypothermia was significantly reduced,and the difference was statistically significant(P<0.01).3.In brain tissue of mir-29cexpression:Compared with model group,acupuncture group,the group,the acupuncture combined hypothermia group at low temperature can reduce the mir-29 c expression in the brain tissue,there are significant difference statistically significant(P<0.01);Compared with acupuncture group and mild temperature group,the expression trend of mir-29 c was significantly decreased in acupuncture combined with mild hypothermia group,but the difference was not statistically significant(P>0.05),which may be related to the lack of sample size.4.Map4k4,Map3k14 and Fem1 b in ischemic side hippocampal tissue:Compared with the model group,the expression of Map4k4 in the brain tissue could be down-regulated by acupuncture group,mild hypothermia group,acupuncture and mild hypothermia group,and the difference was statistically significant(P<0.01).Compared with mild hypothermia group,the expression of Map4k4 could be significantly reduced by acupuncture combined with mild hypothermia,and the difference was statistically significant(P<0.01).Compared with the model group,the expression of Map3k14 in brain tissue could be down-regulated by acupuncture group,mild hypothermia,acupuncture and mild hypothermia,and the difference was statistically significant(P<0.01).Compared with hypothermia group and acupuncture group;The expression trend of Map4k4 was significantly reduced by acupuncture combined with mild hypothermia,but the difference was not statistically significant(P>0.05),which may be related to the lack of sample size.Compared with the model group,the expression of Fem1 b in the brain tissue could be increased by acupuncture group,mild hypothermia,acupuncture and mild hypothermia,and the difference was statistically significant(P<0.01).Compared with acupuncture group and mild hypothermia,Fem1 b expression trend was significantly increased in acupuncture combined with mild hypothermia,but the difference was not statistically significant(P>0.05),which may be related to the lack of sample size.Conclusion:1.the acupuncture group,the group,the acupuncture combined hypothermia group at low temperature can reduce neural function defect scale,reduce the cerebral infarction area ratio,protect brain tissue,especially acupuncture combined hypothermia group.2.The protective effect of each treatment group on brain tissue may be achieved by lowering the expression of mir-29 c and its target genes Map4k4 and Map3k14 on MAPK pathway and up-regulating the expression of mir-29 c target gene Fem1 b.3.in reducing cerebral infarction area ratio,increase the expression of Fem1 b,acupuncture combined the low temperature is better than acupuncture group and the low temperature group,may be because the acupuncture with hypothermia therapy played a role.
