Acanthopanax evodiaefolius Franch belonging to an Araliaceae family is mainly distributed in the Yangtze River Basin and the south of the Yangtze River in China,and its dried roots and stem barks are listed officially in the Chinese Pharmacopoeia(2015 edition)as Acanthopanax Cortex(named as Wujiapi),which has been used as medicine for the treatment of paralysis,arthritis,rheumatism,lameness,and liver disease.In this study,we investigate the effect of water extracts(WZY-WA)? MeOH extracts(WZY-Me)?PE?EA and BU franction(WZY-PE?WZY-EA?WZY-BU)of Acanthopanax evodiaefolius Franch on ?-glucosidase inhibitory activities,?-amylase inhibitory activities,PTP1 B inhibitory activities,DPPH?ABTS radical scavenging assay,cytotoxicity and GSIS assay.Phytochemical investigation on the leaves of Acanthopanax evodiaefolius Franch led to the isolation of twenty-five compounds,and their structures were identified as naringin(1),prunin(2),remain to be identified(3),stigmasterol(4),taraxerol(5),daucosterol(6),remain to be identified(7),scopolin(8),apigenin-7-O-neohesperidin(9),syringin(10),hyperoside(11),clavane-2?,9?-diol(12),7-O-(?-D-Glucosyl)-apigenin(13),maltol(14),4H-1-Benzopyran-4-one,3-(?-D-glucopyranosyloxy)-6,7-dihydroxy-2-(4-hydroxyphenyl)-(15),corosolic(16),(±)-pinoresinol(17),(+)-medioresinol(18),phenol,2,6-dimethoxy-4-[tetrahydro-4-(4-hydroxy-3-methoxyphenyl)-1H,3H-furo[3,4-c]furan-1-yl]-,[1S-(1?,3a?,4?,6a?)]-(9CI)(19),(±)-syringaresinol(20),naringenin(21),maltol-3-O-?-D-Glucoside(22),6,8-Di-C-?-D-glucopyranosylapigenin(23),ethylconiferin(24),ethylsyringin(25),respectively.To our best knowledge,compounds 1,2,5,8~9,11~13,15,17~20,22~24 were first obtained from the leaves of Acanthopanax evodiaefolius Franch.Compounds 1,2,9,12,13,15,19,22~25 were reported for the first time from the Acanthopanax genus.Compounds 1,9,12,13,15,19,22~25 were firstly isolated from the family Araliaceae.To establish a method for simultaneous determination of the contents of eight flavonoid in the leaves of Acanthopanax evodiaefolius Franch by quantitative analysis of HPLC.We found that the contents of eight flavonoid were very distinctive in the leaves of Acanthopanax evodiaefolius Franch in different collecting period.The results shows that the content of naringin were most higer than other flavonoid when the leaves collected in June and October,reached13.13 mg/g and 13.76 mg/g,respectively;the content of eight flavonoid is both relatively low when collected in August and naringin and 7-O-(?-D-Glucosyl)-apigenin were not detected.The results provide the scientific basis for exploiting resources of A.evodiaefolius Franch.To optimize the conditions of the polyamide resin purification of naringin from leaves of A.evodiaefolius Franch,purification procedure of polyamide resin was investigated by response surface methodology.The effects of five factors on purification of naringin were studied.With yield of naringin as index,methanol concentration,elution volume and sample volume were optimized by response surface methodology.The optimized conditions were as follows:methanol concentration of 30%,elution volume was 3.0BV,sample concentration was 4.0mg/mL,sample volume was 3.5BV.Under these conditions,yield of naringin reached 56.12%,which agreed with model prediction.The optimal purification condition of naringin with polyamide resin from A.evodiaefolius Franch was stable and reliable.Moreover,a series related hypoglycemic avtivity test of three compounds that isolated from leaves Acanthopanax evodiaefolius Franch have been investigated.Among tested compounds,corosolic with the concentration of 5 ?M have strong ?-glucosidase inhibition,and it also have a good inhibitory effect on PTP1 B enzyme with low cytotoxicity.Naringin and prunin shows weakly hypoglycemic effects.The results demonstrated that corosolic may possess potential hypoglycemic activity and is worth of further investigation.Hypoglycemic activity of other compounds that isolated from leaves of Acanthopanax evodiaefolius Franch need further research.
