Acanthopanax trifoliatus(Linn.)Merr belongs to the genus of Araliaceae,which is mainly scattered in the central and southern parts of China,it is represented by A.trifoliatus,which is produced in Jiangmen of Guangdong Province and Ganzhou of Jiangxi Province.Take root or root bark,with leaf twigs into the medicine,taste bitter,spicy,slightly cold.It has the effects of blood circulation and relaxation,clearing away heat and detoxification,phlegm and dampness,etc.which has been used as medicine for the treatment of treat chest tightness cough,fever,cold,rheumatism and pain.In this paper,the inhibitory activities ofα-glucosidase from methanol extract(ATR-ME),petroleum ether extract(ATR-PE)and ethyl acetate extract(ATR-EA)of stems of A.trifoliatus were studied.The bioactive sites ofα-glucosidase with good inhibition,low cytotoxicity are ethyl acetate extract(ATS-EA).Inflammation has an important relationship with the pathogenesis of type 2 diabetes.The cytotoxicity and NO、TNF-α、IL-6inhibition of methanol extract(ATS-M),petroleum ether extract(ATS-PE),ethyl acetate extract(ATS-EA)and n-butanol extract(ATS-Bu)from stems of A.trifoliatus were investigated.Ethyl acetate extract(ATS-EA)with low cytotoxicity and better NO inhibition was selected as the bioactive fraction.The activity of the chemical constituents was studied,separation by column chromatography and preparation of liquid phaseand of A.trifoliatus,a total of 21 compounds were isolated and purified.1H-NMR,13C-NMR and DEPT were used,combined with physical andchemicalpropertiestoidentifythestructure,including phthalicacid(1),taraxerol(2),kaurenoicacid(3),stigmasterol(4),taraxerol acetate(5),to be identifified(6),to be identifified(7),β-sitosterol、stigmasterol(8),3α11α-dihydroxy-23-oxo-lup-20(29)-en-28-oicaicd(9),querc etin-3-O-β-D-galactopyraniside(10),acantrifoside A(11),to be identifified(12),pubinernoid A(13),syringin(14),daucosterol(15),stigmasterol glucoside(16),acankoreoside A(17),rutin(18),4-Hydroxymethylbenzaldehyde(19),to be identifified(20),n-Butyl-β-D-fructopyranoside(21).Among them,the compounds10,13,16,19,21werefirstisolatedfromtheA.trifoliatus,Araliaceae and Acanthopanax Miq.The activity ofα-glucosidase inhibitory activity of A.trifoliatus was investigated.It was found that ATS-EA had lower cytotoxicity and higher inhibition rate ofα-glucosidase than positive control acarbose,indicating that ATS-EA has the effect of hypoglycemic.To investigate the cytotoxicity of LPS-induced mouse macrophage cell line RAW264.7 and the inhibition of NO,TNF-αand IL-6 inflammatory factor release,the ATS-EA cytotoxicity was low and the inhibition rate of NO,TNF-αand IL-6 inflammatory factors was higher than that of the positive control Dexa,indicating that the anti-inflammatory effect was better.
