Part 1Effects of PM2.5 on expression of IL-8 and SIRT1 in bronchial epithelial cellsObjectives:To explore the effects of particulate matter(PM)2.5 on the expression of IL-8 and SIRT1 in bronchial epithelial cells.Methods:BEAS-2B cells were subcultured in growing medium until to 80%in fusion,and then starved in serum-free medium for 6 hours.The cells were devided into four groups:control group,PM2.5 25 group,PM2.5 50 group and PM2.5 100 group.Cells in control group were incubated with PM2.5 of 0 ?g/mL.Cells in other groups were treated with a series of concentrations(25,50 and 100 ?g/mL)of PM2.5 for 24 h.Supernatants and cells were harvested in each group.IL-8 level in supernatants was analyzed by ELISA,and SIRTI protein level in BEAS-2B cells was detected by western b ot methods.Results:Compared to the control group,IL-8 levels in the supernatants in other groups were significant increased with a concentration-dependent manner.(all P<0.05),and SIRT1 protein levels in BEAS-2B cells induced by PM2.5 were decreased in concentration-dependent manner(all P<0.05).Conclusions:IL-8 levels were improved and SIRT1 protein expression levels were reduced in BEAS-2B cells induced by PM2.5 with a concentration-dependent manner.Part 2Effects of paeonol on the expression of IL-8 and SIRT1 in bronchial epithelial cells induced by PM2.5Objectives:To investigate the effects of paeonol on the expression of IL-8 and SIRT1 in BEAS-2B cells induced by PM2.5.Methods:BEAS-2B cells were subcultured in growing medium until to 80%in fusion,and then starved in serum-free medium for 6 hours.The cells were devided into six groups:control group,paeonol 15 group,paeonol 30 group,PM2.5 100 group,paeonol 15+PM2.5 100 group and paeonol 30+PM2.5 100 group.Cells in control group were incubated with PM2.5 of 0 ?g/mL.Cells in the paeonol 15 group and paeonol 30 group were treated with paeonol of 15 ?mol/L and 30 ?mol/L respectively.Cells in the PM2.5 100 ?g/mL group was stimulated with PM2.5 of 100 ?g/mL for 24 h.Cells in paeonol 15+PM2.5 100 group and the paeonol 30+PM2.5 100 group were pretreated with paeonol of 15 ?mol/L and 30 ?mol/L respectively for 1 h,and then stimulated with PM2.5 of 100 ?g/mL for 24 h.Supernatants and cells were harvested in each group.The protein expression of IL-8 in the supernatants was analyzed by ELISA.The SIRT1 protein expression in BEAS-2B cells was measured with western blot methods.Results:Compared to the control group,the protein expression levels of IL-8 and SIRT1 had no statistical significance in the paeonol 15 group and paeonol 30 group(all P>0.05).Treatment with paeonol could decrease the IL-8 levels and increase the SIRT1 protein levels in BEAS-2B cells induced by PM2.5 compared with the PM2.5100 group(all P>0.05).The IL-8 level in paeonol 30+PM2.5 group was lower than that in paeonol 15+PM2.5 group(P<0.05),and the SIRT1 protein level in paeonol 30+PM2.5 group was higher than that in paeonol 15+PM2.5 group(P<0.05).Conclusions:Paeonol could inhibit the expression of IL-8,and enhance the expression of SIRT1 protein in BEAS-2B cells induced by PM2.5 in concentration dependent manners.
