Article one:A study of paeonol makes effect on the proliferation rate of blastofibriocyte and mesothelial cellObjective Peritoneal adhesion is one of the common complications postoperatively and its incidence is above 90%.Among those sufferings,more than 80% get adhesion between wounds and epiploon,50%involve intestines.Trauma,especially the operation history,is the most frequently reason of the peritoneal adhesion.Peritoneal adhesion can induce internal organs twist and change,so they cause internal organs dysfunction and persistent pains,which caused by abdominal wall dragged.With resulting in chronic abdominal pain,women infertility,etc.,Peritoneal adhesion is normally related with trauma,inflammation,corpus alienum remained,etc.,in the research,we found that peritoneum's injury is a important priming factor of peritoneal adhesion;the next-to peritoneal injury is the space foundation of constit;the earlier fibra exudation offers important framework for blastofibriocyte increased and finally to form adherence.To decrease the next-to peritoneal injury,promote fibrinolysis in the early stage,facilitate mesothelial cell repairing peritoneum,and inhibit the increase of blastofibriocyte,all of these methods are very important to prevent peritoneal adhesion. To make use of the traditional Chinese medicine Cortex Moutan can repair mesothelial cell and regulate blastofibriocyte's function effectively,so it could lighten peritoneal adhesion.In the clinical medicine practice,the patient with peritoneal adhesion caused by disclosed abdomen operation.It needs ambrosia in a certain time after operation,so before the functional recovery of stomach intestine,we can't give the traditional Chinese medicine orally,and lost the time window of preventing peritoneal adhesion.If a non-give intestinal tract drag can be found,it is very important in clinical application. Paeonol is main effective constituent in Cortex Moutan.In order to test its efficacy,we designed follow experiment.The objective of experiment is to study of paeonol makes effect on the proliferation rate of blastofibriocyte and mesothelial cell.Methods 20 healthy Wistar rats were used to cultivate mesothelial cell and blastofibriocyte.Divide cultured cell into groups,group A(blastofibriocyte),group B(mesothelial cell),group A1 (blastofibriocyte+1640),group A2(blastofibriocyte+LPS),group A3 (blastofibriocyte+TNF),group A4(blastofibriocyte+LPS+paeonol),group A5(blastofibriocyte+TNF+ paeonol),group A6(blastofibriocyte+ paeonol);group B1(mesothelial cell+1640),group B2(mesothelial cell+LPS),group B3(mesothelial cell+TNF),group B4(mesothelial cell+LPS+paeonol),group B5(mesothelial cell+TNF+ paeonol),group B6(mesothelial cell+ paeonol).After one day's cultivated,count its number with MTT and use HE stain to identify the cells.Results Use LPS or TNF alone can motivated the blastofibriocyte proliferation,but when join in the paeonol,it can significantly reduce the blastofibriocyte proliferation which effected by the inflammatory factor LPS and TNF.Use LPS alone can motivated mesothelial cell proliferation,but when join in the paeonol,it reduced proliferation rate of mesothelial cell;Use TNF alone can inhibit mesothelial cell proliferation,but when join in the paeonol,it neutralized the inhibition of TNF,promoted mesothelial cell proliferation and reparation. Conclusion Mesothelial cell and blastofibriocyte are very important cells in the process of peritoneal adhesion.It is believed that when something suscitated peritoneum, Mesothelial cell function degraded and its number decreasd,whereas blastofibriocyte activated,proliferated overload.After the change of cell number,effected by the complicated physiological functionsin in the organism,peritoneal adhesion process started.The traditional Chinese medicine paeonol can directly inhibit blastofibriocyte activation and proliferation,and it also have the up-regulation effects in mesothelial cell proliferation,so it can promote peritoneal reparation in the early stage.Article Two:The modulatory action of Paeonol in tPA & PAI secreted by macrophages and mesothelia cellsObjective Menzies said that serous membrane which caused by mechanical injury,tissue ischemic,corpus alienum motivation and peritonitis etc.were the most remote cause of remote cause.The past research showed that the exudation and coagulation of the hemaleucin is an important link to form peritoneal adhesion.The process of the fibrinolysis was controlled by t-PA/PAI effectively.Early days research had found that t-PA and PAI are mainly secreted by peritoneal mesothelium cell and macrophage,which provides possibility of a non-give intestinal tract drag。The objective of this experiment is in order to study the influence of the adhesions factors secreted by mesothelial cell and macrophage,which paeonol makes effect on.Methods 15 Wistar rats are used culturing macrophages and peritoneal mesothlial cells. Macrophages are in group A,mesothlia cells are in group B.There are four sub-groups in group A.They are macrophages-1640,macrophages-Paeonol,macrophages-LPS, macrophages-LPS- Paeonol separately.There are six sub-groups in group B.They are mesothelia cells-1640,mesothelia cells- LPS,mesothelia cells-TNF,mesothelia cells-LPS- Paeonol,mesothelia cells-macrophages- Paeonol,mesothelia cells- Paeonol separately.After one day culturing,macrophages in supernate fluid are taken to measure the contents of TNF,t-PA and PAl.Results inflammatory factor TNF and LPS can enhance the mesothelia cells' excreting level of t-PA.Associated with Paeonol,excreting level of t-PA in those mesothelia cells activated by TNF is enhanced more,and excreting level of PAI in those cells is reduced.But inflammatory factors are no use to those cells activated by LPS.Only Paeonol is no use excreting t-PA and PAI in mesothelia cells. Conclusion The foundation of peritoneal adhesion is inflammatory factors releasing, malfunction of mesothelia cells,all that result in decreasing of t-PA/PAI ratio,activity of plasmin decreasing,fibrin effusion increasing,and fibric adhesion.The Chinese herb-Paeonol can modulate the excreting of t-PA/PAI effectively by changing the interaction of macrophages and mesothelia cells.It can also promote fibrin dissolving.Thus it can prevent peritoneal adhesion occur.Article three:Model preparing of traumatic peritoneal adhesion in rats and function of Paeonol studyObjective Peritoneal adhesion is normally related with trauma, inflammation,corpus alienum remained,etc.Peritoneal adhesion is one of the common complications postoperatively and its incidence is above 90%.Peritoneal adhesion can induce internal organs twist and change,so they cause internal organs dysfunction and persistent pains,in the early stage,in the ingredient of the oral intake Chinese herds,Rheum and salivia miltiorrhiza can reduce inflammatory reaction so that they help to restrain the adhesion more.The traditional Chinese medicine paeonol can directly inhibit blastofibriocyte activation and proliferation, and it also have the up-regulation effects in mesothelial cell proliferation,which This research aimed at preparing the appropriate animal model to investigate the extent of the above two factors' influence on the peritoneal adhesion.Methods 400 healthy Wistar rats were randomly divided into five groups with eithty animals in each group,each sex in a half respectively.The surgeries on them were different among the different groups:Group A(Trauma group),intestines were clamped to establish an abacterial traumatic model;Group B(Trauma+ Paeonol group), intestines were clamped and Paeonol im.0.2ml/200mg per day;Group C(Taruma+ Sodium Hyaluronate group),the procedures made were the same as those in Group A and lay Sodium Hyaluronate on the traumatic of small intestine 0.2ml;Group D(trauma+Sodium Hyaluronate+Paeonol),on the base of group C,Paeonol im. 0.2ml/200mg per day_postoperatively;_Groop E(Normal control group),without any treatments.Rats in all the above groups were randomly divided into four groups,which were killed at 3 day,7 day,1.4 day,21 day postoperatively in turn to observe their status of peritoneal adhesion.The adhesion tissues were taken to dye in HE.Adhesion classification data were analysed with ANOVA.Results Adhesion status were the most severe in Group A.In general,the degree of adhesion status decreased in the following turn:Group A>Group C>Group B>Group D>Group E.With HE dying it suggested that macrophages and blastofibriocyte increased,mesothelial decreased in the specimen of severe adhesion animals,while it is not notability in the specimen of mild adhesion animals.Conclusion The basic pathophysiological changes in peritoneal adhesion are the release of inflammatory factors,mesothalial's malfunction,the activity of plasmin decreasing,and the increase of fibrin's effusion.Chinese herbs Paeonol can depress the inflammatory reaction,keep back the normal function of mesothalial,increase the level of plasmin.So the effusion of fibrin decreases and adhesion forms less.The modulatory mechanisms of Chinese herbs on the formation of peritoneal adhesion is that it can reduce the production or inflammatory factors and the effusion of fibrin,increase the activity of plasminogen activators.
