Objective:Research astragalus glycosides(ASTIV)and total notoginseng saponin(PNS)compatibility effect on ischemia reperfusion injury in rats,and from the four main effective components in vivo pharmacokinetic behavior to investigate both the synergy to enhance resistance mechanism of cerebral ischemia-reperfusion injuryMethods:Modified line bolt of middle cerebral artery of rats cerebral ischemia model,through observation and ASTIV PNS compatibility to nerve funtion score of rats,cerebral infarction area,the influence of HE staining,the comprehensive evaluation of ASTIV and compatibility of PNS pharmacological effects against cerebral ischemia reperfusion injury;Using high performance liquid chromatography-tandem level 4 pole mass spectrometry determination of compatibility in the plasma of rats different time ASTIV and ginseng saponin Rg1 and ginseng saponin Rb1,the content of notoginseng saponin R1,comparative pharmacokinetic parameters change,investigate the effective component of ASTIV and compatibility of PNS pharmacokinetic behavior changes.Results:ASTIV,PNS and its compatibility to reduce ischemic reperfusion rat infarction area,reduce nerve disfuntion ethology score,alleviate nerve function defect symptoms,improve the pathological morphological changes after cerebral ischemia.ASTIV and PNS compatibility effect is stronger than the two alone.ASTIV and PNS compatibility at the same time,the ASTIV and ginseng saponin Rg1 and ginseng saponin Rb1,panax notoginseng saponins R1 AUC increased significantly,the average residence time lengthen the MRT(0-t),Cmax increased significantly,and in the clearance rate of slow down significantly.ASTIV and PNS compatibility and significantly affected the ASTIV and PNS pharmacokinetic behavior in the body,make the drug increased the absorption of active ingredients in the body,slow down,spend more time,so that the blood drug concentration increasedConclusion:In cerebral ischemia reperfusion rats,ASTIV and PNS compatibility with synergy to enhance resistance to ischemic brain damage.Its mechanism might be ASTIV and compatibility of PNS makes its four main effective components of pharmacokinetic behavior change,thus promote the extent Of drug absorption and speed,extend the drugs in cerebral ischemia model rats,retention time,greatly enhance the bioavailability and to enhance effectiveness and extend the efficacy,play for the purpose of the synergies of cerebral ischemia.The results from the point of pharmacokinetic and pharmacodynamic reveals the effective constituents in radix astragali and notoginseng compatibility rationality for the treatment of cerebral ischemia.
