Pharmacokinetics research and Individual administration design of MTX in cancer patientsAbstract The serum concentration of MTX was monitered after 0,1,2,4,8,12,24,26,28,30,36,48hours of MTX injection on cancer patients (breast cancer and Thyroid gland Carcinoma of thyroid),who were divided into two groups, one group is accompanined with pleural effusion ,the other group is without pleural effusion.The pharmaeokinetis models and parametas of these two groups were investigated, and the individual administration designs were given.This experimentfocused on the following aspects :1. The selection of pharmacokinetics models for these two groups, both are two-compartments models.2. The determination of weights for these two groups, both are 1/C.3.The estimation of pharmacokinetics parameters for these two groups ,the t1/2αof the group with pleural effusion is significently longer than the group without pleural effusion, they are 1.405 ±0.41h and 1.005±0.30h respectively; the t1/2βof the group with pleural effusion is also siginficently longer than the other group , which are 16.26±9.06h and 8.15±1.43h respectively.4.Individual administration designs were given according to the pharmacokinetics parameters.5. The fealibility of individual administration design were verified.The main resulst of thisstudy are :1.The pharmacoknetics parameters for cancer patients who are in different physiological state were obtained.2.The individual administration design was given ,and verified. The new findings of this study were :1. The pharmacokinetics parameters of MTX for cancer patients were determined .2. The individual administration designs were given, which provide guidelines,both theoretical and pratical, for the more rational use of MTX .
