Mechanisms Of Yiqi Jiedu Xiaoying-Prescription On Th17/Treg Immune Imbalance In Rats With Experimental Autoimmune Thyroiditis

Abstract/Summary:
Objective:To observe the regulatory effects of Yiqi Jiedu Xiaoying Formula on the levels of thyroid autoantibodies(TGAb,TPOAb)and cytokines(RORyt,Foxp3,TGF-β;IL-6,IL-10,IL-17)in experimental autoimmune thyroiditis rats(EAT),and to preliminarily explore the regulatory effects of Yiqi Jiedu Xiaoying Formula on Th17/Tre in EAT rats.The mechanism of G immune imbalance provides a reliable experimental basis for the treatment of autoimmune thyroiditis(AIT)with traditional Chinese medicine.Methods:Sixty SPF Wistar rats were selected as normal group,and the remaining 50 rats were modeled..Forty-eight rats were randomly divided into model group,sodium selenite control group,high-dose treatment group of Yiqi Jiedu Xiaoying Formula and low-dose treatment group of Yiqi Jiedu Xiaoying Formula,12 rats in each group.Double steamed water,sodium selenite and Yiqi Jiedu Xiaoying Mixture were given orally for 8 weeks.Peripheral blood regulatory T cells(Treg)and helper T cells 17(Th17)were measured by flow cytometry.At the end of the 10th week,the thyroid cells of anesthetized rats were harvested.The morphological changes of thyroid cells in EAT rats were observed by HE staining.Serum levels of IL-6,IL-17,IL-10 and TGF-beta cytokines in EAT rats were measured by ELISA.The expressions of stat3,RORyt,Foxp3 and STAT5 mRNA in thyroid tissues of rats were detected by real-time quantitative RT-PCR.Results:1.The level of serum thyroid antibody in EAT rats was significantly higher than that in normal rats.After 8 weeks of treatment,the serum thyroid antibody levels of rats in high,low dose and Western medicine groups decreased.The high-dose group of traditional Chinese medicine is better than the western medicine group,and the high-dose group of traditional Chinese medicine has a dose-effect relationship.2.In the normal group,the thyroid follicles were intact,filled with glia,and the follicular epithelium was cubic.No inflammatory cell infiltration was observed.In model group,thyroid follicles were destroyed extensively and the structure was incomplete.A large number of lymphocyte infiltration was seen in the thyroid gland.Compared with the model group,the thyroid gland in the western medicine group improved,some follicular structures recovered and lymphocyte infiltration decreased.Most of the follicular structures recovered and lymphocyte infiltration decreased in the high and low dose groups.The high dose group was better than the low dose group.3.Compared with normal group,Treg in PMBC of EAT rats decreased significantly,Th17 increased significantly,and Th17/Treg ratio increased.Sodium selenite western medicine group and high and low dose Chinese medicine group can significantly increase Treg,reduce Th17 ratio and Th17/Treg ratio.The traditional Chinese medicine group is superior to the western medicine group and has a dose-effect relationship.4.Compared with normal group,the expression of Foxp3 in thyroid gland of EAT rats was lower and the expression of RORyt was higher.After treatment,Foxp3 expression increased and RORγt expression decreased in western medicine group and high and low dose Chinese medicine group.The traditional Chinese medicine group is superior to the western medicine group and has a dose-effect relationship.5.Compared with normal group,serum levels of IL-17,IL-6 and TGF-β in EAT rats increased,while IL-10 decreased.After treatment,the levels of IL-17 and IL-6 decreased,while the levels of IL-10 and TGF-β increased in western medicine group and high and low dose Chinese medicine group.The high-dose group was better than the western medicine group and the low-dose group.6.Compared with the control group,the expression of stat5 and Foxp3 in spleen tissue of the model group was significantly down-regulated;compared with the model group,the expression of stat5 and Foxp3 in the high-dose group and the low-dose group were significantly up-regulated,while the expression of stat5 and Foxp3 in the western medicine group was not significantly up-regulated,and there was no statistical difference;there was significant difference between the traditional Chinese medicine group and the western medicine group.Compared with the control group,the expression of ror gamma T and stat 3 in spleen tissue of model group rats was significantly increased.Compared with model group,the expression of ror gamma T and stat 3 in high-dose group and low-dose group of traditional Chinese medicine were significantly down-regulated.There was significant difference between traditional Chinese medicine group and Western medicine group.ConclusionYiqi Jiedu Xiaoying Decoction can reduce the level of thyroid autoantibodies(TGAb,TPOAb)in EAT rats,improve the thyroid pathological changes in EAT rats,and regulate the Th17/Treg imbalance in EAT rats.The mechanism is to regulate the expression of RORyt and Foxp3 and the levels of cytokines(TGF-β,IL-6,IL-10 and IL-17)in thyroid of EAT rats.
