Licorice is the most used herbal medicine in China since ancient times.With the deep research,it's in resource shortage because of its widely used in many fields in a long term.Now,licorice Pieces mainly from Neimeng,Gansu and Xinjiang.Gansu is th e earliest documented licorice's habitat area which has multiple licorice planting base.In recent years,Guazhou County has become the largest licorice planting base.Extraction is an important part of production.Now there is a low extraction effic iency and monitoring lag issues in pharmaceutical industry because of progressive scal e-up of extraction technology and moniter rely on experience.So we chose Ural licoric e of Gansu's Guazhou as the research object,and carried out the research of extraction optimiation.For study,the index component method of Gansu licorice were establishe d,licorice's extraction technology was optimized and amplified.In order to provides met hods and theoretical for the extraction process,the on-line analytical method for pilot s cale licorice extraction process was established,and the extraction process was studied i n depth by fitting the kinetic equation.The main findings are as follows;1 The establishment of index components content determination method:Establish an HPLC method for the determination of liquiritin,isoliquiritin,liquiritig enin and glycyrrhizic acid.Establish,and a UV method for the determination of total lie orice flavonoids in roots and knotty rhizome of G.uralensis.The methodological study showed that a good linear correlation(r>0.9990),precision,reproducibility,stability,av erage recovery(<3%).It can serve as a method for the determination of five index co mponents of licorice.Eight batches of roots and knotty rhizome of licorice are determi ned,and the average content of five index component in roots of licorice were 0.513%,0.0729%,0.0484%,1.945%,2.19%,while they were 0.456%,0.06 3 6%,0.036%,1.630%,2.578%in knotty rhizome of licorice.It is provide a reference for promotion of the knotty rhizome of licorice.2 Optimization of licorice's extraction technology:The extraction technology of licorice was studied using heat reflux method with t he content of five index component and paste forming rate as the index.Levels of fa ctors were determined by single factor experiment of extraction solvent,material-solve nt ratio,extraction time and extraction times.The optimal technology of licorice was e xtracted 1.5h three times with an 8-fold amount of water by orthogonal experiment.T he small test demonstrated the transfer rate of five constituents all reaching over 70%.The pilot test result showed that the transfer rate and extraction yield of the five act ive constituents were all basically stable,demonstrating that the technology conditions can be adapted to pilot scale production.3 The establishment of licorice's NIRS online detection method:Based on different bathes of licorice's pilot test,NIRS samples were collected onli ne.The optimal pretreatment methods was original spectrum by comparing the model's PRESS,with the optimal modeling band by SiPLS methed.The calibration model of li quiritin,isoliquiritin,glycyrrhizic acid and total licorice flavonoids was built by PLS method.The verification result showed the predicted value of the model has a good c orrelation with the true value,which can be use to monitor the changes during the lic orice extraction process,and lay a foundation of online quality control of licorice extr action.4 The preliminary study of licorice's extraction dynamicsThe content of index component and extraction time were fitted by empirical mo deling method under the optimal extraction technology.The result shows that the Higu chi equation was the optimal quation for every process of licorice's extraction with a high correlation coefficient.The higuchi equation which simple described the licorice's extraction was related to its structure,provided a theoretical reference for further resea rch and amplify.
