Objective:Metabolic Syndrome(MS)includes dyslipidemia,elevated blood pressure,and elevated plasma glucose,it is closely tied to cardiovascular diseases.The basic character of MS can be summarised as"central obesity,dyslipidemia,hypertension,and insulin resitace".In recent years,MS is highly pravalent and became a serious medical problem which affects people's health and quality of life.In this study,Stagnation of liver-Qi with deficiency of the spleen is considered to be the cause of MS in traditional chinese medicine(TCM)theory,and effects of Chai-Qi decoction on NF-?B passway is the main outcome measures,to explore the effect of Chai-Qi decoction on MS and its mechanism.Methods:8-week old rats were divided into normal group(NG),model group(MG),preventive chinese medication group(PCMG)and chinese medication group(CMG).NG received a regular diet of rat chow.and other groups received a diet with high fat,sugar and salt.Chai-Qi decoction was started to give to PCMG at the 1st week,and to CMG at 9th week.sICAM-1 and sVCAM-1 were examined,and also the expression of NF-?B and I?KPwere examined at the end of 4th week,8th week and 12th week.Abdominal aortic sections were observed under transmission electron microscope.Results:1.ELISA1.lsICAM-1:(1)4th week:the sICAM-1 concentration was significantly higher in MG than NG.(P<0.01),PCMG is lower than MG(P>0.05)but higher than NG(P<0.01),(2)8th week:MG is higher than NG(P<0.01).PCMG is lower than MG(P<0.05)but higher than NG(P<0.05).12th week:MG is higher than NG(P<0.01),PCMG is lower than MG(P<0.01),CMG is lower than MG(P>0.05),both CMG and PCMG are higher than NG(P<0.05),CMG is higher than PCMG(P<0.05)1.2 sVCAM-1:(1)4th week:the sVCAM-1 concentration was significantly higher in MG than NG.(P<0.01),PCMG is lower than MG(P>0.05)but higher than NG(P>0.05).(2)8th week:MG is higher than NG(P<0.01).PCMG is lower than MG(P>0.05)but higher than NG(P>0.05).12th-week:MG is higher than NG(P<0.01),PCMG is lower than MG(P<0.01),CMG is lower than MG(P>0.05),CMG is higher than NG(P<0.01),PCMG is higher than NG(P>0.05),CMG is higher than PCMG(P<0.05)2.Western BlottingThe expression of NF-?B and I?K? in MG are higher than NG at the 8th week and the 12th week,PCMG is higher than NG but lower than MG;at the 12th week,the expression of I?KP is lower in CMG than PCMG,but NF-?B is higher in CMG than PCMG;the expression of NF-?B and I?K? in MG are lower than NG at 4th week,both are higher in CMG than NG.3.Abdominal aortic sectionsCompared with the normal group,smooth muscle cells and the collagen fiber increased in model group,platelet components,macrophage,endothelial cell swelling and vacuolar degeneration were also observed.CMG shows less Pathological changes above than MG,and cell proliferation is lower in CMG than MGConclusions:1.Chai-Qi decoction possessed the protective effects on abdominal aortic vascular endothelial injury of MS rats.2.Chai-Qi decoction attenuates adhesion molecule expression by the regulation of NF-?B passway.3.The effects of early intervention of Chai-Qi decoction is better than treatment,and eliminating the cause of MS should be the best mean.
