Depleted uranium(DU),a waste product of uranium enrichment,has several civilian andmilitary applications.It was used as armor-piercing ammunition in international military conflicts due to its availability,price and pyrophoricity.and was claimed to contribute to health problems,known as the Gulf War Syndrome and recently as the Balkan Syndrome.Inhaled depleted uranium(DU)aerosols are recognised as a major human health hazard.Depending on aerosol speciation,inhalation may lead to a protracted exposure of the lung and other organs.However,a desirable agent to clear DU has not been discovered until now.In this experiment,a canine model of lung injury induced by depleted uranium inhalation had been setup,which setted up by two methods,and then whole lung lavage(WLL)had been operated in those models,in order to observe the therapeutic role of WLL to DU induced lung injury.The experiment is divided into two sections.Part 1:To establish a canine model of lung injury induced by depleted uranium inhalation.The objective is to establish a canine model of lung injury induced by depleted uranium inhalation.Twelve dogs were randomized into three group:normal-control group(n=4),Suspension-polluted group(n=4),air sac-polluted group(n=4)They were polluted by DU with injecting suspension(80mg/kg)through tracheal intubation,or with dust(10mg/kg)by air sac.The CT were observed in the first and third and ninth day.,and the urine as well as the blood were collected in the reasonable time for the inspection of liver or kidney’s function.And the urine content were also surveyed.At lastwhen they died naturally,the pathological examination of lung tissues were observed and noted.We found that CT scanning revealed patchy and effusion in left lung in Suspension-polluted group.And no injury in sac-polluted group.Uranium level of the urine in group air sac-polluted was higher than the suspension-polluted group.And also the former is severitier than the after in the function of liver and hepar.Pathologically,changes were noted in lung tissue of Suspension-polluted group,such as escape of inflammatory cells into alveoli,hemorrhage and dilation and congestion of alveolar capillaries,and infiltration of inflammatory cells in interstitial tissue.But in the air sac-polluted group,pathological changes were found significantly,such as hyperplasia and fibrosis,tubercle.The two methods could introduce the canine model of lung injury induced by depleted uranium inhalation.Part 2:To observed the treatment of whole lung lavage with dogs of lung injury induced by depleted uranium inhalation.The object is to observe the treatment of whole lung lavage with dogs of lung injury induced by depleted uranium inhalation.Twelve dogs were randomized into three group:DU-polluted group(n=4);Saline-lavage group(n=4);CPE-lavage group(n=4).The dogs were given whole lung lavage when 48 hours afted inhaled DU dust and the collect the bronchoalveolar lavage fluid for the uranium content.The CT were observed in the day lavaged and seventh day,and the urine as well as the blood were collected in the reasonable time for the inspection of liver or kidney’s function.And the urine content were also surveyed.At lastwhen they died naturally,the pathological examination of lung tissues were observed and noted.Results:CT scanning revealed the DU dust of lung and trachea disappear at the first day and the eighth day with lavage.Uranium level of the urine in the CPE lavage group was lower than the Saline lavage group,and the two lavage group were higher than the DU-polluted group.And the Saline group is severitier than the CPE group in the function of liver and hepar.In conclusion,the whole lung lavage is effective in treating lung polluted with DU.
