Objective:The purpose is to investigate the effect of Yes-associated protein(YAP)gene silencing by siRNA on the Biological behavior of human hepatoma cancer cells HepG2.Methods:In our experiment,cell transfection method was used to transfect YAP siRNA into hepatoma cells HepG2.The expression of YAP mRNA and protein were detected by RT-PCR,Western blotting and immunofluorescence confocal;Cell viability was detected by morphological observation,MTT assay and colony formation assay;Scratch experiment was used to detect the effect of the migration of HepG2 cells.Cell apoptosis was detected by Flow cytometry and the expression of cell apoptosis related proteins were investigated by western blot analysisResult:The expression of YAP mRNA and protein was inhibited significantly.The experimental group HepG2 cells became round,even detached from the culture flask.MTT test and colony formation assay have shown that the silencing of YAP gene inhibited the growth of HepG2 cells significantly.Cell scratch test showed the cell migration became slower.The percentage of cell apoptotic was increased from 6.12%to 34.62%.The result of Western blotting showed that caspase-3 and Bcl-2 were decreased significantly,while Bax had no change.Furthermore,the expression of Cox-2 was decreased.Conclusion:Use siRNA technique,the specific YAP siRNA may down regulate the expression of YAP gene in HepG2 cells.It can inhibit the proliferation,migration and promote the apoptosis of HepG2 cells.
