Objects To explore the expressions of thioredoxin reductase(TrxR) mRNA and protein, and the relationship with TrxR and myeloid leukemia, for myeloid leukaemia diagnosis, effect and prognosis provides experimental basis.Methods60samples of patients with bone marrow, including20AML,20CML, and20samples of healthy men as normal control group were collected, Selection of human non small cell lung cancer A549cells as the positive control group. The expression of TrxR mRNA was detected by real-time quantitative polymerase chain reaction(real-time PCR), The expression of TrxR protein level was detected by Western Blot.Results The results of Real-time PCR showed that the expression levels of TrxR mRNA in AML, CML and A549group were significantly higher than normal controls(X2=18.392, P=0.000,P<0.001); For AML patients, the expression levels of TrxR mRNA in the primary diagnosed and relapse, treatment, A549group were significantly higher than controls (X2=35.160,P=0.000,P<0.001), the difference between the primary diagnosed and relapse group and A549cells was no significant (P=0.981,P>0.05), but significantly higher than CR group(P<0.001), the expression of CR group compared with controls were higher(P?0.001); For CML patients, the expression levels of TrxR mRNA in the primary diagnosed, treatment, A549group were significantly higher than controls(X2=38.22,P=0.000,P<0.001), the difference between the primary diagnosed and A549group was no significant(P=1.000, P?0.05), but significantly higher than treatment group(P<0.001), the expression of treatment group compared with controls was significantly higher(P<0.001); The expression of TrxR mRNA in the primary diagnosed and relapse group of AML and the primary diagnosed group of CML showed no significant difference(P=0.989,P>0.05). The result of Western Blot showed that the expression of TrxR protein in AML, CML and A549group were significantly higher than the normal control group.Conclusions The result of experiment show that the TrxR gene and protein expression level increased in myeloid leukemia. At the mRNA level, the expression level of TrxR gene in primary diagnosed and relapse group were increased than CR group in AML, the expression of CR group compared with controls were significant differences; the expression level of TrxR mRNA in CR group are significantly lower than control group; The expression level of TrxR mRNA in primary diagnosed group than treatment group increased significantly In CML, the expression of treatment group compared with controls was significant differences; The expression of TrxR mRNA in the primary diagnosed and relapse group of AML and the primary diagnosed group of CML showed no significant difference; In TrxR protein expression level, the expression of TrxR in myeloid leukemia higher than control group. The results showed that, TrxR in myeloid leukemia cell proliferation process may play an important role. The low expression of TrxR in normal hematopoietic stem cells were increased in myeloid leukemia, and after treatment, the expression level of TrxR mRNA were decreased, which can be used as one of the parameters to predict the incidence,effect and prognosis of myeloid leukemia.
