First part: To investigate the expression of nuclear factor erythroid-2 related factor 2 (Nrf2)gene in chronic myeloid leukemia(CML)and explore its relationship with clinical characteristicand the correlation with thioredoxin reductase (TrxR). The expressions of Nrf2 and TrxR geneswere analyzed in 30 bone marrow prepares of CML patients in different phases, including 20 inchronic phase(CML-CP),3 in accelerated phase(CML-AP),7 in blastic phase(CML-BP) andK562 cells using real-time quantitative polymerase chain reaction(real-time PCR).Ten healthysubjects were served as normal controls.The results reveal that the expression of Nrf2 mRNA inAP,BP group showed no significant difference,but lower than that of CP group. Nrf2 gene ishighly expressed in the first visit group of CML than other groups,and is decreased aftertherapy,which may be the molecular marker predicting the progress of CML.Nrf2 mRNAexpression level is correlated with TrxR.Second part: To further study the correlation between the expressions of Nrf2 gene andTrxR. On the basis of the principle of target sequence of siRNA,four interference sequences ofNrf2 were designed and linked into lentivirus carrier.The Nrf2-RNAi-LV interference sequenceswas transfected into K562 cells ,the mRNA level of Nrf2 and TrxR genes were analyzed byreal-time PCR.The results reveal that the expression of Nrf2 mRNA in experimental group 3was effectively downregulated,whereas the cells in negative control group had not obviouslychanged, the expressions in negative control group and blank group showed no significantdifference. It hints that Nrf2 may affect TrxR expression through Nrf2-ARE signal way ,thenplay influence in apoptosis of cell,which provides an experimental basis for research about therole of Nrf2-ARE signal way in leukemia and may provide advice in target therapy with CMLusing RNAi.
