OBJECTIVE:This study studied the germplasm resources of Bletilla and the quality standards of Bletilla seed seedlings,selected excellent germplasm for seed production of Bletilla,established Bletilla seed seedling production technical regulations and Bletilla seeds.Seedling quality standard technical system.It is the original source of medicinalBletilla,regulates the seed seedling production of Bletilla,and provides experimental basis and technical support for Bletilla's standardized planting.METHOD:?1?Bletilla's germplasm evaluation16 Bletilla germplasm resources were collected,and the genetic diversity was detected at the molecular level by SRAP molecular marker method.Thepolysaccharides of Bletilla,the gelatin and mineral elements of Bletilla were determined,and the HPLC fingerprint of Bletilla germplasm resources wasconstructed.Correlation analysis,principal component analysis,cluster analysis and other statistical methods were used to study the germplasm resources of Bletilla.?2?Bletilla's seed quality standard studyThe seed production process,such as seed formation,seed maturity,seed harvesting time,and seed storage methods,as well as capsule morphology,seedmorphology,seed vigor,and seed germination rate,were studied in Bletilla's capsules and capsules.?3?Study on the quality standards of Bletilla seedlingsThe agronomic traits and grades combined action of multiple factors.The order of influence is origin>breeding mode>variety>growth period.The HPLC fingerprint of Bletilla germplasm resources was constructed.The 16Bletilla resources had 20 common peaks.The peak areas of the common components of Bletilla germplasm resources of different varieties and cultivation methods were clustered by means of the mean method.Analysis,found that the presence andabsence of Bletilla ingredients and the level of content affected by the variety,cultivation methods,production areas.?2?Bletilla's seed quality standard studyBletilla's appearance?length,width,weight?is significantly related to seedmaturity.Seeds with high maturity can be screened by measuring the appearance of capsules.Seeds with large volume and heavy weight have third-class capsule or the deformed and defective capsule is the outer capsule;Bletilla's seed moisture content is the key factor forBletilla seed storage,and temperature is the second important factor for Bletilla seed storage.The seed moisture content was reduced to 5%and stored at 4°C as the best storage condition for Bletilla seeds.?3?Study on the quality standards of Bletilla conclusion:?1?SRAP molecular markers,mineral element content,and HPLC fingerprinting can be used for the evaluation of Bletilla germplasm resources.SRAP molecularmarkers can be used to evaluate the genetic diversity of Bletilla germplasm resources and the identification of authenticity.Minerals The elemental content can be used to evaluate the quality of Bletilla's germplasm resources,and the HPLC fingerprint can be used to identify the authenticity of Bletilla's germplasm resources.?2?Through Beckilla's capsule and seed formation process,appearancemorphology and seed maturity,capsule grading,and seed storage methods,Bletilla's seed production technical regulations and Bletilla's seed quality standard technical system were established.?3?Through the research on the appearance of seedlings of three types of Bletilla,the field life after transplanting,and establish the technical system of Bletilla seedling quality standard.
