Bletilla striata was belonged to campanulaceae codonopsis perennation, and had high ornamental and medical value.In this paper, the tissue culture and rapid propagation of Bletilla striata (Thunb.) Reichb. F.were explored.The wild seed was served as experimental materials, and TTC stain method was used to explore the effect of different storge on seed vigor. The influence of experimental factor, including 6-BA,NAA,TDZ, and snail enzyme concentration and processing time, on seed sterile germination was researched. The effect of three plant growth regulator (NAA,TDZ,6-BA) on multiplication proliferation and differentiation was studied, while the influence of different medium, active carbon, and NAA on root. The three plant growth regulators (NAA,TDZ,6-BA) on the proliferation and differentiation of the original bulbs analyzed by orhthogonal design. At the same time, the plant growth regulator NAA and different concentrations of activated carbon with different basic medium was used on rooting growth.Bletilla seedlings was experienced on the survival rate and growth by the different of substrate. A、B、C、F strains of separation from roots was inoculated on base material,measured the data of the survival rate of the test tube seedlings and the growth rate of the height of the measurement. The experimental results are as follows:(1)With the increasing time of storage,storage time for 7d,30d,60d seeds have high energy, but the seed vigor in lowering, seed vigor is respectively 95.2%, 94.3% and 93.5%.(2) The best culture medium is MS+1.5 mg/L 6-BA+0.5 mg/L NAA+0.03 mg/L TDZ,the germination rate can reach 91.6%,the experimental phenomenon is growth and tidy. At 25 degree with 40 min, the processing and concentration of 3% snail enzyme, seed germination rate is highest, which can up to 93.4%.The experimental results shows that the strain of A, B, C and F significantly improve seed germination rate, the strain of A、B、C and F 23.5%、22.6%、29.7% and 28.9% respectively. Treated by strain C,the experimental phenomenon is a small amount of second leaves, which could promote the growth of seeds.(3)The optimal medium for proliferation of protocorm is MS+0.4 mg/L NAA+ 0.9mg/L6-BA+0.06 mg/L TDZ,the rate is up to 123.4%.TDZ is used in the experient is better than the other hormone NAA and 6-BA. The optimum culture medium for the differentiation of protocorm is MS+0.6 mg/L 6-BA+0.2 mg/L TDZ+1.5 mg/L NAA; the rate is up to 69.4%.the rootage of the experiment is 1/2MS+0.05%AC+ 0.5 mg/L NAA, the rate is up to 97.3%。(4) Transplanting 30ds,the survival rate of tube seedlings were similar, all can achieve 93.6% above, but the plant growth status difference is very big, with matrix is bark, wood shavings, sawdust for 3:6:1, leaf, stem straight, growing exuberant;blade stretching is poorer, plant growing weaker. Strain A, B, C, F respectively with transplanting seedlings, the survival rates were similar, strain, strain, strain C,B,A strain F in 45 d transplanting survival rate were 64.44%,68.89%,66.67%,62.22%.After 30ds,the difference of the survival rate of plantlets is not habe too large difference.All of them could reach more than 93.6%.but it has differen phenmenon of the growth status of plants.The best of them is with matrix bark, shavings and sawdust at 3:6:1.he survival rate of plantle, which deal with A, B, C and F strains can reached 64.44%,68.89%,66.67%,62.22%.
